This book is devoted to alkaliphiles, their microbiology, biotechnological applications and adaptive mechanisms. Alkaliphiles are extremophilic organisms that are adapted to thrive in alkaline environments. Over the years, a wide variety of alkaliphiles belonging to domain Bacteria, Archaea and Eukarya have been isolated and studied. These organisms use various adaptive mechanisms to thrive in ‘extreme’ alkaline environments, and some of these adaptive mechanisms are of immense importance to a range of biotechnological applications. In this book, readers will learn about the adaptive strategies of alkaliphiles in colonizing alkaline habitats, with a main focus on: (1) the production of enzymes that are active and stable in the high pH environment, and (2) the production of acids that decrease the pH of their immediate surrounding environment. Enzymes that are operationally stable at high pH (also known as alkaline active enzymes) are desirable in several applications such as detergent formulating and leather tanning processes, and they are among the major selling enzymes and the most important industrial enzymes. The growing demand in many existing and emerging biotechnological applications led to the discovery, characterization, engineering and evaluation of diverse types of alkaline active enzymes. In addition to the use of these fascinating enzymes in biotechnological applications, readers will discover the mechanisms of action and stability of these enzymes at extreme pH. Studies have shown that some alkaliphiles decrease the severity of the high pH of their media by producing substantial amount of organic acids, which could be of great interest in various applications presented in this book. In addition to enzymes and organic acids, other products of biotechnological importance such as carotenoids, bioactive substances, and chelators have also attracted researchers’ attention. Whole-cells of alkaliphiles have been used as food and feed, and are also useful in environmental applications such as in waste treatment and construction.

This book presents the fundamentals of the reservoir and interfacial engineering. The book systematically starts with the basics of primary, secondary and tertiary (enhanced) oil recovery and emphasizes on the theory of microbial-enhanced oil recovery (MEOR) and its potential toward recovery of oil in place. Different approaches of MEOR such as in-situ, ex-situ, and integration of chemical- and microbial-enhanced oil recovery (EOR) are discussed in detail. This book highlights the link between the effectiveness of MEOR and the local reservoir conditions, crude oil characteristics, and indigenous microbial community. The latest implementations of MEOR across the globe are highlighted as case studies to outline the potential as well as the scope of MEOR. Given the topics covered, this book will be useful for professionals and researchers working in the areas of petroleum science and engineering, chemical engineering, biotechnology, bioengineering, and other related fields.

Enzymes are nature’s biocatalysts empowered with high catalytic power and remarkable substrate specificity. Enzymes perform a wide range of functions throughout nature, and guide the biochemistry of life with great precision. The majority of enzymes perform under conditions considered normal for mesophilic, neutrophilic, terrestrial microorganisms. However, the Earth’s biosphere contains several regions that are extreme in comparison, such as hypersaline lakes and pools, hydrothermal vents, cold oceans, dry deserts and areas exposed to intensive radiation. These areas are inhabited by a large number of extremophilic microorganisms which produce enzymes capable of functioning in unusual conditions. There is an increasing biotechnological and industrial demand for enzymes stable and functioning in harsh conditions, and over the past decade screening for, isolation and production of enzymes with unique and extreme properties has become one of the foremost areas of biotechnology research. The development of advanced molecular biology tools has facilitated the quest for production of enzymes with optimized and extreme features. These tools include large-scale screening for potential genes using metagenomics, engineering of enzymes using computational techniques and site-directed mutagenesis and molecular evolution techniques. The goal of this Research Topic is to present reports on latest advances in enzymes from all types of extreme environments. Contributions dealing with isolation of enzymes from extremophilic microorganisms or directly from natural environments, screening for and expression of enzymes with extreme properties using metagenomic approaches are welcome. In addition, contributions dealing with all forms of biocatalyst production and improvement are welcome, such as fermentation technology, protein engineering, directed evolution, rational design, and immobilization techniques.
