Since the apoE4 allele is a risk factor or susceptibility gene in late-onset familial and sporadic AD, the mechanism of disease expression may involve metabolic effects that are isoform specific. Isoform-specific interactions of apoE therefore become critical in the mechanism of AD pathogenesis. Detailed characterization of the binding of the apoE isoforms with proteins and peptides relevant to the pathology of the disease may be critical in understanding disease pathogenesis. These critical isoform-specific interactions of apoE may involve interactions with proteins and pep tides in the defining neuropathologic lesions of the disease, the neurofibrillary tangle and senile plaque. Other possible critical isoform-specific interactions include the mechanism of internalization, intracellular trafficking, and subsequent metabolism. In addition, differential post-translational modifications of apoE isoforms may determine differences in metabolism contributing to the pathogenesis of the disease. Oxidation of apoE may confer several isoform-specific, biochemically distinct properties. Since {3A peptide binds apoE in the lipoprotein binding domain of the protein and not in the receptor-binding domain, apoE could target bound {3A4 peptide to neurons via the LRP receptor. Internalization of the apoEI {3A peptide complex into the cell, by the same route as the apoE-containing lipoproteins, would result in incorporation into primary lysosomes and pH dependent dissociation. The demonstration of apoE in the cytoplasm of neurons, with isoform-specific interactions of apoE with the microtubule-binding protein tau demonstrated in vitro, suggest additional, testable hypotheses of disease pathogenesis.

"Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by cognitive decline and ultimately death. The primary pathological hallmark of (AD) is the presence of amyloid beta protein which can accumulate intracellularly and eventually aggregate into extracellular plaques. Amyloid beta has a variety of toxic effects and is particularly disruptive to the brain's cholinergic system, a neural network essential for learning and memory function. In this study, the McGill THY-1-APP mouse model of AD, which overexpresses the precursor of amyloid beta, has been used to profile the region and time-specific impact of amyloid pathology on cholinergic synaptic integrity. As a point of comparison, the impact of amyloid pathology on the glutamatergic system has also been investigated. Finally, in an effort to protect the cholinergic system from amyloid beta toxicity, a matrix metalloproteinase 9 inhibitor (MMP9i) has been administered to aged THY-1-APP transgenics and their respective C56Bl/6 wild type controls. This study was unable to confirm the cholinergic early up-regulation hypothesis which postulates that there is a compensatory increase in cholinergic structure and function prior to extensive amyloidosis and plaque deposition. However, this study was able to confirm end-stage cholinergic synaptic losses and the relative resilience of the glutamatergic system to amyloid toxicity. At the present time, whether or not administration of the MMP9i is capable of preventing end-stage cholinergic losses remains unclear as the results are inconclusive. On the one hand, with the relatively small sample sizes used, there is some preliminary evidence to suggest that administration has a protective effect. This is because the density of cholinergic boutons in transgenic THY-1-APP MMP9i-treated mice did not differ significantly from wild type C57Bl/6 vehicle-treated mice, while the density of cholinergic boutons was significantly lower in vehicle-treated THY-1-APP transgenic mice as compared to wild type C57Bl/6 vehicle-treated mice. On the other hand, the THY-1-APP transgenic MMP9i treated mice failed to attain a statistically significant elevation in cholinergic bouton density when compared directly with the THY-1-APP transgenic vehicle treated mice, suggesting that the drug was unable to prevent cholinergic bouton loss after all. Further testing will be required in animal models with a larger sample size and less variance before firm conclusions can be drawn as to the status of the MMP9i's putative protective ability. Finally, the MMP9 inhibitor failed to improve the performance of treated wild type C57Bl/6 mice or THY-1-APP transgenic mice on a behavioural test for episodic-like memory. The author concludes that the MMP9i is worthy of future investigation as a therapeutic agent and that if one hopes to correct cognitive dysfunction then co-administration of other therapeutic agents may also be necessary." --
