Abstract: Abstract of thesis entitled ANTI-OBESITY EFFECT OF BITTER MELON (MOMORDICA CHARANTIA) Submitted by Qixuan Chen for the degree of Doctor of Philosophy at The University of Hong Kong in December 2004 The overall objective of this study was to test the hypothesis that bitter melon (Momordica charantia) possesses anti-obesity properties. Increased adiposity is known to trigger insulin resistance, and the onset of metabolic syndrome. The public has substantial interest in dietary adjuncts that could reduce the risk of obesity and diabetes. Bitter melon is the most popular herbal resource for lowering blood glucose, and its hypoglycemic effect is well documented, although the mechanism has not been elucidated. In the present study, a series of experiments were performed to test the hypothesis that the hypoglycemic effect of freeze-dried bitter melon (BM) juice on rats is associated with a reduction in adiposity. Sprague-Dawley rats were given a low-fat (LF, 7%, w/w) or a high-fat (HF, 30%) diet for 6 weeks. At the beginning of week 7, the LF-diet fed rats either continued to consume the LF-diet or were switched to a LF+1.5%BM diet. HF rats continued to consume the HF-diet or were switched to a BM supplemented diet (0.375%, 0.75%, or 1.5%). At the end of week 15, HF-diet fed rats gained more weight and had higher visceral fat mass compared to the LF group. BM supplementation at a dose of 0.75% or above had improved glucose tolerance. Momordica charantia L. is growing in many tropical and subtropical regions; the fruits of bitter melon are also gradually becoming popular for treating diabetes and associated diseases. Over 248 compounds belonging to the lipids, phenolics and terpenoids class are reported by diverse studies. However, M. charantia L. appears to be an inimitable species that synthesizes a diverse range of natural products in the fruits, leaves, stems and roots. The cucurbitane types of triterpenes exist in the various tissues of the plant in their aglycone as well as glycosylated forms. The bitter melon seems to exert their lipid lowering and antiobesity effects via several mechanisms like PPARs, LXRs, SREBPs, and Sirts mediated fat metabolism in various tissues, prevent adipocyte hypertrophy and visceral fat accumulation. M. charantia L. has been comprehensively studied worldwide for its therapeutic properties to treat a number of diseases like diabetes, dyslipidaemia, obesity, and certain cancers. This chapter apparently displays an encompassing literature review on vast potential of bitter melon as antiobesity agent and assembles data on complex phytochemistry.
