As obesity is a significant public health concern, strategies to improve control over appetite and body weight are in high demand. There is some evidence to suggest that long-chain polyunsaturated fatty acids (LCPUFA) may have greater appetitive suppressing effects than shorter-chain, mono- and saturated fatty acids, but the literature surrounding the effect of n-3 LCPUFA on appetite, body weight and body composition is inconsistent. The overall aim of the following work was to investigate whether n-3 LCPUFA could reduce appetite and body weight more effectively than shorter-chain, less saturated fatty acids. A series of investigations were undertaken. The first was a meta-analysis to examine the strength of evidence for an effect of n-3 fish oil on body weight and BMI. Following the development of orosensory matched oil-based emulsions (containing predominantly docosahexaenoic acid (DHA), or oleic acid (OA), or a blend of oils that reflect the Western diet (WB)), a twelve-week, randomised, controlled intervention was conducted to examine the effect of consuming a daily dose of DHA or OA on energy intake, body weight, BMI and body composition. Two randomised, controlled, acute feeding studies were then conducted to examine the effect of consuming OA or DHA on energy intake and OA, DHA or WB on subjective appetite scores and energy intake. Finally, an exploration into the potential mechanisms by which DHA could exert an anorectic effect, and an evaluation of saliva as a potential source of appetite biomarkers, was conducted. The results of this investigation do not provide consistent evidence of the potential of n-3 LCPUFA to reduce appetite or EI, but this may be due to the limitations of measuring appetite with the available tools. Proteomic analysis of saliva samples taken during one of the acute feeding studies identified specific protein alterations associated with reduced energy intake. Preliminary investigations into relationship between salivary proteins and feeding behaviour suggests there is further value in examining saliva and targets identified as potential surrogate indicators of appetite. The identification of novel biomarkers that complement or replace existing measures of appetite could help elucidate the potential effects of n-3 long-chain PUFA on appetite and body weight.
