The presence of monotypism in thick atherosclerotic lesions of black females with G-6-PD mosaicism first reported by the Benditts (1973) has been confirmed in two other laboratories. However, we believe that it is premature to conclude that the finding of monotypism necessarily indicates monoclonal origin of athero sclerotic lesions. We have suggested two alternative explanations for the obser vation of monotypism which we believe must be shown to be invalid before accept ing monoclonal origin as the only plausible way to account for the observed G-6-PD monotypism. One of these two alternatives relates to clonal heterogeneity of cell growth potential, i. e. , during the course of progressive growth of a le sion, progeny of one cell may overgrow all others in a portion of the lesion. The other alternative is that one of the G-6-PD alleles may be linked to genes that afford a preferential survival characteristic in the abnormal environment present in atheroscerotic lesions. Thus, cells with one allele may be able to grow better than cells with the other allele, and this characteristic may be unrelated to "A-ness" or "B-ness". We have studied initiation of lesions in He diet-fed swine and demonstrated that all active lesions that were studied were of multiple cell origin (not monoclo nal). We have studied cell growth patterns in developing atherosclerotic lesions in He diet-fed swine and found evidence consistent with clonal heterogeneity in growth potential of lesion cells.

The Advanced Study Institute on Regression of Atherosclerotic Lesions, sponsored by the North Atlantic Treaty Organization, was held in Maratea, Italy, September 11-13, 1983. The institute was designed to foster discussion of experimental data on the regression of atherosclerosis in animals and evidence of a similar phenomenon in humans. The participating scientists showed with anatomical and biochemical data, that atherosclerosis regresses in cholesterol fed animals after withdrawal of atherogenic stimuli. The existence of regression in humans was inferred from sequential angiographic visualization of arteries, but anatomical and biochemical characteristics of the process have not been defined. Two mechanisms are probably involved in such an angiographic regression. The first, which may occur when hyperlipidemia is partly or fully corrected or during undernutrition or other metabolic abnormalities, may be related to atherosclerosis changes noted in animals undergoing regression. The second type seems independent of changes in plasma lipid levels and may involve resolution of thrombi or other underlying arterial wall factors. The relative frequencies of the two mechanisms are unknown; whether they can occur simultaneously also warrants consideration. It is likely that an exciting milestone will be forthcoming when regression of atherosclerosis is unequivocally demonstrated in humans. Publication of these institute proceedings constitutes a valuable background for future developments.

Low density lipoproteins (LDL) are pathophysiologically important be cause of their central role in the disease atherosclerosis and because atherosclerosis is the leading cause of death in developed countries. Many researchers believe that a more detailed knowledge of the struc ture, function, and metabolism of LDL may eventually lead to a means to control atherosclerosis. For this reason a fairly large research effort has gone into the investigation of LDL over the past few years. The purpose of this book is to collect and summarize in one place most of the pub lished information on LDL through 1975. To this end more than 1500 references are cited in the papers that make up this volume. The A, B, C apolipoprotein classification system was adopted for use throughout this work. In addition to the A, B, C, and "D" families of apolipoproteins, apoE is used to designate the "arginine-rich" apolipo protein. This classification system is used because it is far less cumber some than other proposed classification schemes for apolipoproteins.
