The NATO Advanced Study Institute on Biomechanics of Active Movement and Division of Cells was held September 19-29, 1993 in Istanbul and the Proceedings are presented in this volume. Sixty-eight scientists from sixteen countries attended. Prof. J. Bereiter-Hahn of Goethe-Universitat, Frankfurt, Germany, Prof. A.K. Harris of the University of North Carolina, Chapel Hill, USA, Prof. R.M. Nerem of Georgia Institute of Technology, Atlanta, USA and Prof. R. Skalak of the University of California, San Diego, USA were the members of the International Organizing Committee. As the Scientific Director of the Institute, I wish to express my sincere appreciation for their assistance without which the Institute could not have taken place. This Institute is the third one of the meetings which are now called "the NATO Istanbul Meetings on Cytomechanics". The first one was the NATO Advanced Research Workshop on Biomechanics of Cell Division which was held October 12-17, 1986 in Istanbul. The Proceedings were published as NATO ASI Series A Life Sciences Vol. 132 by Plenum Press in 1987. The second one was the NATO Advanced Study Institute on Biomechanics of Active Movement and Deformation of Cells which was held September 3-13, 1989 in Istanbul. The Proceedings were published as NATO ASI Series H : Cell Biology Vol. 42 by Springer-Verlag in 1990.

Cytomechanics is the application of the classical principles of mechanics in cell biology. It is an applied science concerned with the description and evaluation of mechanical properties of cells and their organelles as well as of the forces exerted by them. Thus, this topic needs a truly interdisciplinary approach, and accordingly this volume gives an up-to-date account of the current research done on cell division, mitosis, cytokinesis, cell locomotion and cell deformation during normal development and the cytoskeletal role in cell shape. Biologists, biomechanicians, biophysicists, biochemists and biomathematicians here discuss the basic concepts of mechanics and thermodynamics, emphasizing their applicability to cell activities.

This volume contains the lectures presented at the NATO Advanced Study Institute (ASI) on "Trafficking of Intracellular Membranes: From Molecular Sorting to Membrane Fusion", held in Espinho, Portugal, from June 19 to June 30,1994. The objective of this Institute was to survey recent developments and to discuss future directions in the rapidly advancing field of membrane cell biology, with particular emphasis on the dynamical properties and intracellular flow of membranes. A wide range of interrelated topics around the central theme of intracellular trafficking of membranes was covered, including lipid flow, membrane fusion, dynamics of membrane components, protein folding and assembly, vesicular transport in membrane biogenesis, exocytosis and endocytosis. A large variety of experimental techniques and systems, including the application of viruses and model systems, to study these processes was also considered. Membrane cell biology is a broad discipline which encompasses many scientific areas including cell biology, biochemistry, biophysics, virology, immunonology and genetics. Indeed, recent advances in the cell biology of membranes could not have been made without this multidisciplinary approach. Significant progress achieved during the last few years in understanding how newly synthesized lipids and proteins find their way to the cell organelles, how molecular sorting and the continuous flow of membranes allow each cellular membrane to maintain its own distinct molecular composition, and, thereby, the individuality of the various intracellular compartments, was discussed in considerable detail in this Institute.

During the last years much attention has been given to the investigation of the roles of specific growth modulators acting on liver cell populations. HGF functions as a pleotropic factor in the liver as well as in other extrahepatic tissues. The hepatic stem cell compartment is thought to play a major role in liver tissue restoration. Mechanisms of cell growth and cell death of the liver are described in detail, with particular emphasis on apoptosis. The functions of HBV-pX protein and the roles of hepatitis B virus gene expression relation to hepatocellular carcinoma are discussed. Techniques for obtaining human and nonhuman hepatocytes and other liver cell types and for efficient transfer and expression of genes in hepatocytes and liver-derived cell lines are presented.

This is now the fourth time that protein phosphorylation has been the focus of a NATO Advanced Study Institute. The first meeting with the topic "Signal Trans duction and Protein Phosphorylation" was held on the island of Spezai, Greece, in September 1986. The second one took place in Chateau La Londe, France, in September 1989 on "Cellular Regulation by Protein Phosphorylation", the third one on " Tyrosine Phosphorylation/Dephosphorylation and Downstream Signaling" was in September 1992 in Maratea, Italy. The titles of these books clearly mirror the developments that have taken place in the last decade. Beginning with the recognition that protein phosphorylation is at the center of signaling -clearly established in 1990 -it became apparent that many cellular processes are regulated by this mode. A new focus then emerged when it was recognized that growth factors are bound to corresponding receptors trigger protein tyrosine phosphorylation which controls cell prolifera tion. This was the topic of the third meeting in this series. It is now evident that further progress depends on understanding the three dimensional structure of the proteins involved. It goes without saying, for example, that understanding the location of proteins by adaptor proteins is only possible on the basis of the three dimensional protein structure. Therefore, the fourth meeting in this series concentrated on the protein structure of signaling molecules as well as on the elucidation of the principles of protein domain interactions.

Even though transmitter release and re-uptake are very complementary and physically in close proximity, researchers usually focus on only one of these two aspects. This volume, however, covers both processes and brings together two major presynaptic events in transcellular communication. It comprises 24 chapters and is divided into two parts: Part I, Neurotransmitter Release, and Part II, Neurotransmitter Uptake. The included topics range from ultrastructure and molecular control mechanisms to pathophysiology of some diseased states. Neuronal as well as non-neuronal mechanisms are treated. The concluding chapter emphasizes the importance of interdisciplinary collaboration and provides a link between the events in the presynaptic terminal and behavior.

Provided here are the latest techniques of NMR as applied to the study of proteins, carbohydrates and nucleic acids. The first chapters are devoted to an introduction to NMR and parameters related to molecular structure and molecular interactions. NMR experiments from basic 1D to 2D, 3D and 4D, used in combination with isotopically labelled molecules, are described and a general strategy is presented for biomacromolecular structure determination. Subsequent chapters deal with more advanced principles and techniques and their applications to structural and dynamic processes involving biomacromolecules in solution. Advanced results on peptide, protein, oligosaccharide and nucleic acid structure and recognition are presented.

Gene therapy has generated enormous expectations as it holds a great promise, namely to revolutionize clinical and molecular medicine. This is because the successful introduction of a gene sequence in somatic sells followed by long-term expression at therapeutic levels can fundamentally correct a variety of human disease such as inherited and acquired disorders, cancer and AIDS. The proceedings of the first ASI on Gene Therapy that took place from August 17-28, 1997 on the island of Spetsai, Greece, summarizes recent advances in the field as presented elegantly by the intemationally recognized experts that served as faculty at the meeting. Their contributions constitute brief but comprehensive reviews presented as independent chapters in this book and emphasize the influence of chromatin structure on gene expression, describe the principal mechanisms of gene expression, followed by articles on epigenic elements, viral vectors, non-viral vectors as gene delivery systems, animal models of human diseases, inducible regulatory systems, gene therapy of cancer, AIDS, monogenic disorders and ongoing clinical trials. Gene therapy is the logical progression to human health management because it offers a potential low cost and efficient treatment. Gene therapy can reach it's full potential and satisfy all expectations. However, a number of critical technology advancements need to be completed. These include the development of vectors capable of safe and efficient gene transfer, the control of cell type targeting and regulation of trans gene expression levels.
