T cells play a vital role mediating adaptive immunity, a specific acquired resistance to an infectious agent produced by the introduction of an antigen. There are a variety of T cell types with different functions. They are called T cells, because they are derived from the thymus gland. This volume discusses how T cells are regulated through the operation of signaling mechanisms. Topics covered include positive and negative selection, early events in T cell receptor engagement, and various T cell subsets.

All of the parasitic organisms highlighted in this new book represent medically important human pathogens that contribute significantly to the global burden of disease. As such there is intense interest in understanding the molecular basis of infection by these pathogens—not only with regard to their clinical relevance but also the fascinating biology they reveal. For most of the parasites discussed here the ability to penetrate biological barriers and/or to establish intracellular residence is critical to survival of the pathogen in the mammalian hosts. For other parasites, a tissue invasive phenotype is a key virulence determinant. In the ensuing 18 chapters, select members of this diverse set of protozoan parasites, as well as some examples of the extremely reduced fungal parasites classified as Microsporidia, are discussed within the context of the fascinating molecular strategies employed by these organisms to migrate across biological barriers and to establish residence within target host cells.

In 1980, the IVth International Cyclic Nucleotide Conference was held in Brussels. As this meeting attracted many investigators involved in cyclic nucleotides and calcium role in intracellular regulation, it was thought that this opportunity could be used to organize, prior to the Congress, an in-depth introductory course on the subject. This was carried out as a NATO Advanced Study Institute. The participants included Ph. D. students and M. D. s engaged in a research training, but also fully trained and well known researchers who wanted a refresher course on the whole subject. During the course, most of the participants and lecturers asked to be provided with a text summarizing the basic lectures of the course. This book was therefore conceived as a basic textbook on the regulation and action of intracellular signal molecules, concentrating mainly on cyclic nucleotides and calcium. It was deliberately kept at a basic level. We would therefore be happy if it could be used as an introduction for interested M. D. s or Ph. D. s working in other fields or entering this field and as a general refresher for researchers interested in the subject. For this reason, very general schemes have been asked of the authors, along with reading lists of available reviews rather than extensive bibliographies. The editors should like to thank the NATO Scientific Affairs Committee for having supported the course and Mrs. Gh. Wilmes who prepared the manuscripts. S. SWILLENS J. E.

The biological membranes of cellular organization enfold an important group of membrane proteins called the ATPases, which are not only versatile in maintaining chemical gradient and electrical potential across the membrane but also bring metabolites necessary for cell metabolism and drive out toxins, waste products and solutes that otherwise can curb cell functions. ATPases are distributed virtually in all live forms starting from unicellular to multicellular and also in viruses. There are different types of ATPases, which differ in function and structure and in the type of ions they transport. The three main types of the ion pump ATPase family are: (i) P-type ATPases that transport different ions across membranes and Ca2+ATPases belongs to this catagory (ii) F-type ATPase in mitochondria, chloroplasts and bacterial plasma membranes produce ATP using the proton gradient; and (iii) V-type ATPase catalyzes ATP hydrolysis to transport solutes and maintains acidic pH in organelles like lysosomes. Genetic defects in either of the ATPases cause several diseases and a number of researches have demonstrated the involvement of the members of ATPases in the cell pathology and diseases, thereby penetrating exciting new areas of our understanding. In this book, the authors summarize recent knowledge about the molecular mechanisms associated with Ca2+-ATPase, V-ATPase and F-ATPase in intracellular and extracellular Ca2+ transport, mitochondrial ATP synthase, vesicular H+ transport, and lysosomal pH regulation. This book thereby bridges the gap between fundamental research and biomedical and pharmaceutical applications. The book provides an informative resource to improve ATPase research and modern therapeutic approaches toward different life threatening diseases that are associated with dysregulation of the ATPases.

Current Topics in Cellular Regulation: Volume 31 is a collection of papers that deals with the role of calcium as intracellular messenger, calcium release or entry across the plasma membrane, calcium in neurotransmitter release, and the regulation of free calmodulin levels in neurons by neuromodulin. Other papers discuss the calcium/calmodulin-dependent protein kinase II, the regulation of muscular calcium channels, and the Na+-Ca2+ exchangers. One paper describes the mechanisms of intracellular calcium release or entry in which agonists acting through the phosphoinositide system, or possibly by metabolites or secondary messenger system, can regulate the mechanisms of cellular Ca2+. Another paper describes different approaches in evaluating the possible mechanisms of calcium in neurotransmitter release. The paper notes that imaging techniques can be used in the timing and order of events in the release process. Possible identification by the investigator of specific "releasing" regions at the membranes, such as active zones, can also define more precisely the exact localization of the process. One paper discusses the interaction between released calmodulin and cytoskeleton proteins that can affect polymerization and cross-linking of cytoskeleton polymers, that leads to filopodia formation or extension. The collection will prove beneficial to molecular biologists, general biologists, microbiologists, biochemists, and researchers involved in cellular biology.

This book provides descriptions of experimental methods in research on the cytoskeleton and its relationships to signaling and cell regulation. Thus, it bridges two active and fertile areas of research. The focus is directed particularly towards methods which take advantage of recent advances in molecular biology, microscopy and immunological assays. A second emphasis is on methods for understanding dynamic changes in cells. A third emphasis is on the formation and turnover of macromolecular and supramolecular complexes, which are so important in driving cell regulation and the behaviour of cytoskeletal elements. A combination of practical advice and detailed protocols should make this book valuable for both novice and experienced workers in these burgeoning fields.
