This detailed treatise is written for chemists who are not NMR spectroscopists but who wish to use carbon-13 NMR spectroscopy. It shows why measurement of carbon-13 NMR is needed and explains how the method can - or should - be used for rapid characterization of flavonoids, one of the most diverse and widespread groups of natural constituents. The ftavonoid pigments, one of the most numerous and widespread groups of natural constituents, are ofimportance and interest to a wide variety ofphysical and biological scientists and work on their chemistry, occurrence, natural distribution and biological function continues unabated. In 1975, a mono graph covering their chemistry and biochemistry was published by Chapman and Hall under our editors hip entitled The Flavonoids. The considerable success of this publication indicated that it filled an important place in the scientific literature with its comprehensive coverage of these fascinating and versatile plant substances. The present volume is intended to update that earlier work and provide a detailed review of progress in the ftavonoid field during the years 1975 to 1980. Although cross references are made to The Flavonoids, this supplement is entirely self-contained and where necessary, tabular da ta from the earlier volume are incJuded and expanded here. The choice oftopics in Recent Advances has been dictated by the developments that have occurred in ftavonoid research since 1975, so that not all subjects covered in The Flavonoids are reviewed again here. A major advance in ftavonoid separation has been the app1ication ofhigh performance liquid chromatography (HPLC) and this is reviewed inter alia in the opening chapter on separation techniques. An equally important development in the spectral analysis of ftavonoids has been the measurement of carbon-13 NMR spectra and this subject is authoritatively discussed in Chapter 2 and is also illustrated with the spectra of 125 representative ftavonoids.

This book was developed from the proceedings of the 2nd North American Tan nin Conference held in Houghton, Michigan, June, 1991. The objective of this con ference was to bring together people with a common interest in plant polyphenols and to promote interdisciplinary interactions that will lead to a bet ter understand ing of the importance of these substances. Another objective of this conference was to extend the 'tannin family' by making special efforts to encourage participation by scientists outside the United States, obtain more coverage of the hydrolyzable tannins, and further broaden the scope of coverage from the initial concentration on forestry and forest products. Com parison of the contents of this book with 'Chemistry and Significance of Condensed Tannins' that resulted from the proceedings of the 1st North American Tannin Conference shows the degree that these objectives were met. In developing the second conference, care was taken to assure that this book extends rather than duplicates the coverage of the first conference. Therefore, the two books should be taken together to obtain an up to date coverage of the broad area of chemistry and significance of plant polyphenols. Our thanks go to the authors who so kindly contributed chapters and so pa tiently responded to our requests. Since the late 1950's when proton n. m. r. spectroscopy was first used in organic natural products studies the technique has increasingly con of this important area of chemistry. tributed to the rapid advancement Although the potential utility of 13C n. m. r. was recognized very early, essentially no application of 13C n. m. r. appeared in the literature prior to 1966 and 95% of the existing data are less than five years old. The initially slow growth had its cause in inadequate instrumentation, insufficient sensitivity being the main obstacle. This situation drastically changed with the advent and commercial availability of broadband ex citation and Fourier transform methods, giving natural-abundance 13C n. m. r. and its numerous chemical applications a tremendous impetus. Today BC spectra can be recorded on sample quantities down to the submilligram level, which until recently even withstood proton n. m. r. Paralleling the development of experimental techniques considerable progress has also been made on an understanding of spectral parameters, in particular their stereochemical implications on natural products. Although the large majority of data present up to now deals with known structures, an adequate basis now exists which allows the chemist to use the technique for tackling real problems on unknown molecules.

About 1958, the late Professor R. E. ALSTON and Professor B. L. TURNER, both of the Department ofBotany, The University ofTexas at Austin, initiated a general systematic investigation ofthe legurne genus Baptisia. They found that flavonoid patterns, as revealed by two-dimensional paper chromatography, were valid criteria for the recognition of the Baptisia species and for the documentation of their numerous natural hybrids. Later, they showed that the flavonoid chemistry could be used for the analysis of gene flow among populations. At that time no attempt was made to even partially identify the flavonoids which were detected chromatographically. Neverthe1ess, it soon became apparent that the full value of the chemical data for systematic purposes required knowledge of the structures of the flavonoids. In 1962, one of us (T.J.M.) in collaboration with Drs. ALSTON and TURNER beg an the chemical analysis of the more than 60 flavonoids which had been chromatographi cally detected in the 16 Baptisia species. In the intervening years, a number of chemists and botanists, inc1uding Drs. K. BAETCKE, B. BREHM, M. CRANMER, D. HORNE, J. KAGAN, B. KROSCHEWSKY, J. MCCLURE, H. RÖSLER, and J. WALLACE, participated in the devel opment of techniques and procedures for the rapid identification of known flavonoids and in the structure determination of new flavonoids. In addition, the flavonoid chem istry of many plants other than Baptisia was investigated.
