The proceedings of a workshop conference are presented in this volume entitled Hypothalamic Peptide Hormones and Pituitary Regulation. The workshop was held in Wilson Hall on the campus of the National Institutes of Health, Bethesda, Maryland, during the days of November 1-2, 1976, and is the most recent of three symposia on neuroendocrinology that have been sponsored by the National Institutes of Health. The first one was held on December 6 - 8, 1961, in the New Everglades Hotel at Miami, Florida. During the first meeting, much emphasis was given to the anatomical and physiological basis for the fledgling science of neuroendocrinology. The proceedings of that symposium were published under the title of Advances in Neuroendocrinology, A. V. Nalbandov (ed. ), University of Illinois Press, Urbana, Illinois, 1963. The second workshop was held on January 8 -11, 1969, in the Arizona Inn at Tucson, Arizona, and was unique in several respects. It was evident to the participants that definitive identification and the determination of the chemical structure of at least one hypothalamic releasing factor was at hand (see Workshop Conference on Bioassay and Chemistry of the Hypophysio tropic Hormones of the Hypothalamus: ~Critical Evaluatioi'i':-J. Meites, ed. , The Williams and Wilkins Co. , Baltimore, Maryland, 1970). Much of what was presented at the second workshop was dedicated to methods of bioassay of the various releasing factors.

The pituitary, albeit a small gland, is known as the "master gland" of the endocrine system and contributes to a wide spectrum of disorders, diseases, and syndromes. Since the publication of the second edition of The Pituitary, in 2002, there have been major advances in the molecular biology research of pituitary hormone production and action and there is now a better understanding of the pathogenesis of pituitary tumors and clinical syndromes resulting in perturbation of pituitary function. There have also been major advances in the clinical management of pituitary disorders. Medical researchers and practitioners now better understand the morbidity and mortality associated with pituitary hormone hyposecretion and hypersecretion. Newly developed drugs, and improved methods of delivering established drugs, are allowing better medical management of acromegaly and prolactinoma. These developments have improved the worldwide consensus around the definition of a "cure" for pituitary disease, especially hormone hypersecretion, and hence will improve the success or lack of success of various forms of therapy. It is therefore time for a new edition of The Pituitary. The third edition will continue to be divided into sections that summarize normal hypothalamic-pituitary development and function, hypothalamic-pituitary failure, and pituitary tumors; additional sections will describe pituitary disease in systemic disorders and diagnostic procedures, including imaging, assessment of the eyes, and biochemical testing. The first chapter will be completely new – placing a much greater emphasis on physiology and pathogenesis. Two new chapters will be added on the Radiation and Non-surgical Management of the Pituitary and Other Pituitary Lesions. Cellular Regulation of Secretion and Release is a compilation of papers that deals with the processes in cellular perception of stimuli and the response to them. Part I describes the receptor occupancy and regulation of stimulus through the use of video image intensification techniques. This part also explains a model for the transmembrane regulation of adenylate cyclase that shows the mechanisms that cells use in sensing and responding to extracellular conditions. Part II discusses the early responses of secretory cells including changes in phospholipid metabolism, in electrophysiological events, and in macromolecular carboxymethylation. Physiologic compartmentalization effectively isolates the central nervous system from the rest of the body. This isolation not only provides protection of its delicate function from aberrant peripheral influences but also impedes its diagnostic evaluation.
