"Fibulin-4 belongs to the fibulin family of extracellular matrix proteins, consisting of seven known members and characterized by tandem repeats of calcium-binding epidermal growth factor (cbEGF)-like motifs and a C-terminal fibulin module. Fibulin-4 knockout (-/-) mice die perinatally due to aortic rupture. Electron microscopic investigation of the aortic wall shows severe disruption of the formation of the elastic laminae and little association of the elastic fibers with the vascular smooth muscle cell surface. In humans, genetic mutations in the fibulin-4 gene have been identified in two cutis laxa (loose skin) patients, both of which exhibited aortic aneurysms. In this study, the effect of the dosage of fibulin-4 was investigated with available mouse models. Using fibulin-4 heterozygous (+/-) and knockout (-/-) mice, as well as, fibulin-4 knockdown (+/R and R/R) mice, with reduced levels of fibulin-4, electron microscopic analyses of the aortic wall were conducted. Our results indicate that reduced levels of fibulin-4 lead to elastic fiber and collagen fibril abnormalities. Although a significant amount of elastin is produced in mice with low levels of fibulin-4, the elastic laminae that are formed show a lace-like appearance. Both +/R and R/R mice also show large, irregular collagen fibrils in the aorta, suggesting that fibulin-4 plays an important role in the regulation of collagen fibril diameter. This is a novel finding. In addition, a stoichiometric relationship between fibulin-4 and fibulin-5 was investigated using two available mouse models, Fbln4SMKO: Fbln5+/- and Fbln4SMKO: Fbln5-/-; electron microscopy of the aortic wall was conducted. These mice harbor a reduction in fibulin-4 that is specific to the blood vessels. It was demonstrated that the absence of fibulin-5 in the Fbln4SMKO: Fbln5-/- mice, the lace-like appearance of the elastic laminae was improved. Taken together, these results indicate that a stoichiometric relationship between fibulin-4 and fibulin-5 exist during elastic fiber assembly. " --

Continued and systematic analysis of the mechanics of flexible fibre assemblies dates from about 1945, although the growth of research into textiles after 1920 had included studies of fabric structure and the measurement of mechanical properties. The subject is thus a young one, although this NATO Advanced Study Institute is a sign of developing maturity. However there is an earlier tradition. All this, and much more such as Max Born's doctoral thesis, was brought to our attention in a delightful discourse by Milos Konopasek, who is not only fascinated by the mathematics of Euler and the modern movement of the solutions of bending curves from two dimensions into three by the use of the computer, but also feels a personal link through having lived and studied within sight of the scene of Euler's triumphs in St. Petersburg.

Elastin is a polymeric protein of the extracellular matrix that imparts the characteristics of extensibility and elastic recoil to tissues. Recombinant polypeptides based on the domain structures and sequences of human elastin self-assemble into organized fibrous structures, with physical properties similar to those of native polymeric elastin. Elastin self-assembly is initiated by a temperature-induced phase separation, called coacervation. Previous to this work, coacervation temperature had been the only parameter available to measure propensity for self-assembly. A variety of techniques were developed using spectrophotometry, microscopy, and rheometry to differentiate the stages of self-assembly, thereby enabling independent observation and quantitation of each stage, and allowing investigations into properties of polypeptides and solution conditions affecting these stages.Self-assembly in the presence of non-elastin, matrix-associated proteins showed that these proteins maintained the coacervate as small droplets, which sometimes flocculated into fibre-like structures. Rheometry demonstrated a second temperature-induced transition above the coacervation temperature, which resulted in gelation and viscoelastic characteristics similar to microgels.Kinetic analysis of self-assembly yielded two additional parameters: coacervation velocity and maturation velocity. Examining the effects of agitation, salt concentration, temperature, polypeptide concentration, size of a polypeptide, hydrophobic domain sequence, and cross-linking domain structure on the kinetics demonstrated that coacervation and maturation are independent stages of self-assembly involving distinct mechanisms. Microscopic observations showed that protein-rich droplets of coacervate grew by coalescence to a stable droplet size, which correlated to differences in maturation velocities between polypeptides. Coacervate droplet growth appeared limited by the formation of organized polypeptide at the surface of the droplets, decreasing surface fluidity. Many of the general principles of the physical chemistry of colloids and emulsions appeared to apply to the formation, growth and stabilization of coacervates of the elastin-like polypeptides.Together, these observations have resulted in a greater level of understanding of the entire self-assembly process, and provided a comprehensive model of elastin-like polypeptide self-assembly that relates to in vivo assembly of elastic fibres.
