Chromatin and Chromosome Structure consists of chapters that reflect a 1975 seminar course and the reported observations after the session. The seminar is organized for the benefit of both faculty and students in the Biology Ph.D. Program of the City University of New York. This book begins with a presentation of the conformational studies of histones. It then describes the histone-DNA interactions and the subunits isolated either from chromatin or from nuclei. It also explains the post-synthetic modifications of histone structure. Furthermore, the book talks about the chemistry and function of nuclear nonhistone proteins, low molecular weight nuclear RNA, and polytene chromosome structure. The regulation of gene expression in chick oviduct model system and the main features of lampbrush chromosomes are shown as well. Aside from the students and faculty of the above-mentioned university, this compilation will be invaluable as well to other undergraduate and graduate students; professors; and researchers. It will also be a good reference to those who are specializing in chromatin and chromosome studies.

Integrating classical knowledge of chromosome organisation with recent molecular and functional findings, this book presents an up-to-date view of chromosome organisation and function for advanced undergraduate students studying genetics. The organisation and behaviour of chromosomes is central to genetics and the equal segregation of genes and chromosomes into daughter cells at cell division is vital. This text aims to provide a clear and straightforward explanation of these complex processes. Following a brief historical introduction, the text covers the topics of cell cycle dynamics and DNA replication; mitosis and meiosis; the organisation of DNA into chromatin; the arrangement of chromosomes in interphase; euchromatin and heterochromatin; nucleolus organisers; centromeres and telomeres; lampbrush and polytene chromosomes; chromosomes and evolution; chromosomes and disease, and artificial chromosomes. Topics are illustrated with examples from a wide variety of organisms, including fungi, plants, invertebrates and vertebrates. This book will be valuable resource for plant, animal and human geneticists and cell biologists. Originally a zoologist, Adrian Sumner has spent over 25 years studying human and other mammalian chromosomes with the Medical Research Council (UK). One of the pioneers of chromosome banding, he has used electron microscopy and immunofluorescence to study chromosome organisation and function, and latterly has studied factors involved in chromosome separation at mitosis. Adrian is an Associate Editor of the journal Chromosome Research, acts as a consultant biologist and is also Chair of the Committee of the International Chromosome Conferences. The most up-to-date overview of chromosomes in all their forms. Introduces cutting-edge topics such as artificial chromosomes and studies of telomere biology. Describes the methods used to study chromosomes. The perfect complement to Turner.

It was at the end of the 19th century that a Swiss biologist, Karl Nageli first proposed the existence of hereditary organelles that carried information from parent to offspring. Ensuing decades experienced vigorous studies that led to the development of discovery that chromosomes are indeed the carriers of genetic information. Subsequent studies, especially by Morgan and Bridges, established unequivocally the chromosome theory of inheritance. Today, the structure of chromosome is well established. At the physical level, eukaryotic chromosomes are composed of a single, linear, double helix of DNA. The elementary helical structure involves nucleosomes, comprised of histones around which the DNA is wrapped. A hierarchy of higher order of chromosomal architecture may possibly be responsible for the regulation of gene helical structures expression. The localized condensations of DNA constitute chromomeres. Uncoiled structures sometimes extending from chromomeres, which form loops, is the result of discontinuities in the regular coiling of the DNA in the chromosome.

Contemporary views on the structure and function of chromatin are presented and the history of the development of these ideas as well as the nature of the nucleic acid and protein components of chromatin are reviewed. The structure of chromatin is studied at several levels, and its modes of transcription and replication are analyzed. Chromatin provides researchers with a critical evaluation of current knowledge. It combines much information that has never before been assembled, and evaluates and interrelates it in a critical way. This has not been done before so that readers are not only provided with an overview, but with extensive references to the literature (there are about 2000 references in all).

This volume of Methods in Enzymology includes up-to-date procedures used for the assembly of nucleosomes, chromatin, and nuclei, extending the classical procedures described nearly ten years ago in Volume 170 (Nucleosomes) of this series, and should assist in the further investigation of the ways in which the structural dynamics of chromatin contribute to geh regulation of transcription, replication, recombination, and repair. Also described in this volume are assay for the structure and function of in vitro reconstituted chromatin and for defining the organization and characteristics of natural chromosomal material from yeast (Saccharomyces cerevisae), flies (Drosophila melanogaster), and frogs (Xenopus laevis), as well as from mammalian tissues. The purification and assay procedures for various chromatin remodeling activities, including histone acetyltransferases, histone deacetylases, and SWI/SNF ATPases are detailed. The critically acclaimed laboratory standard for more than forty years, Methods in Enzymology is one of the most highly respected publications in the field of biochemistry. Since 1955, each volume has been eagerly awaited, frequently consulted, and praised by researchers and reviewers alike. Now with more than 300 volumes (all of them still in print), the series contains much material still relevant today--truly an essential publication for researchers in all fields of life sciences.
