The immune system harbors great potential for controlling and eliminating tumors. Recent developments in the field of immuno-oncology has led to unprecedented clinical benefits for a broad spectrum of solid tumors. However, immunotherapy (IT) approaches currently have several limitations including (i) low response rate; (ii) development of resistance and (iii) causing severe immune-related adverse effects (IrAEs), which underline the importance of adequate patient selection. Importantly, IT holds promising synergistic potential when combined with standard-of-care chemotherapy, radiotherapy (RT) and anti-angiogenic therapy (AAT) as part of multi-modal oncologic treatment regimes. Published data suggest that there are potential synergy between RT and AAT, which ultimately could help potentiate the response to IT. However, the complex interactions between RT and IT and/or AAT remain poorly understood. Many research questions including optimal timing, scheduling and dosing, as well as patient selection and side effects of combined therapy approaches, remain to be addressed. This Research Topic aims to give a comprehensive overview of the current field with particular emphasis on the future outlook of RT and AAT as complementary approaches to improve IT in solid tumors.

Delivery Technologies for Immuno-Oncology: Volume 1: Delivery Strategies and Engineering Technologies in Cancer Immunotherapy examines the challenges of delivering immuno-oncology therapies. Oncolytic viruses (OVs) have emerged as a promising anticancer treatment. OVs selectively infect, replicate in, and kill tumor cells. Oncolytic viral therapy occurs in two phases: an initial phase where the virus mediates direct oncolysis of tumor cells, and a second phase where an induced post-oncolytic immune response continues to mediate tumor destruction and retards progression of the disease. For a long time, the therapeutic efficacy was thought to depend mainly on the direct viral oncolysis based on their tumor selective replication and killing activities. But the post-oncolytic anti-tumor activity induced by the OV therapy is also a key factor for an efficient therapeutic activity. Cancer metastasis is the leading cause of cancer-related deaths. Despite the development of anti-cancer drugs over nearly a century, the 5-year survival rate for patients with metastatic cancer, especially those with distal metastases, remains low. Cancer stem cells (CSCs), also known as cancer-initiating cells, refer to a small population of cells in cancerous tissue with unlimited proliferative potential, capable of reconstituting tumorigenesis. CSCs maintain a relatively stable proportion in cancer tissues through self-renewal and differentiation and are likely to tolerate radiotherapy and chemotherapy drugs. When exposed to certain stimulating factors, CSCs rapidly proliferate, leading to drug resistance and cancer recurrence. Importantly, CSCs can be transferred to distant sites through epithelial-to-mesenchymal transition (EMT) and immune escape, resulting in cancer metastasis. Thus, CSCs constitute underlying factors in tumorigenesis, drug resistance, recurrence, and metastasis, making them potential intervention points for the development of anti-cancer therapeutic strategies.
