The dual role of complement in both cancer development and treatment has been investigated extensively and is characterized by a substantial literature that documents the conditions in which complement can either enhance tumor growth or promote the killing of malignant cells. Indeed, there are now numerous examples of monoclonal antibodies (mAbs) that have either been approved by the FDA or that are under active investigation that make use of complement when eliminating cancer cells. Although the direct in vitro killing of mAb-opsonized cancer cell lines by complement-dependent cytotoxicity (CDC) can be readily demonstrated, there are considerable challenges related to the translation of these findings to the clinic, and numerous strategies have been employed to maximize mAb-mediated CDC in cancer treatment. These approaches include the redesign of mAb dosing schedules; engineering the Fc regions of the mAbs to enhance complement activation; treatment with cocktails of mAbs that bind to several different sites on the targeted cells and thus that potentially synergize CDC promotion; and neutralizing the complement control proteins on malignant cells to weaken their defenses against complement. Target sites on malignant cells that have been successfully exploited for mAb-induced CDC include CD20, CD37, CD38, CD52, and Epidermal Growth Factor Receptors. MAbs specific to complement components have served as powerful analytical reagents to investigate the detailed mechanisms of CDC, and they have been employed to document complement activation by cancer cells and to examine the role of complement proteins (in particular C1q and fragments of C3 and C5) in supporting tumor growth. The use of polyclonal and mAb reagents has revealed a role for the intracellular complement system in cancer biology and strategies that focus on the interaction of complement with the tumor microenvironment, and examining the impact of the complotype on the response to immunotherapy in cancer should lead to additional mAb-based therapies. Along these lines, there is now increasing evidence that strategies that make use of mAbs or other agents to modulate the action of C3a/C5a or their respective receptors may also find use in cancer immunotherapy.

In general, a humoral immune response to breast cancer cells is not therapeutically effective, likely due, at least in part, to the overexpression of membrane bound complement inhibitory proteins on tumor cells. In these studies we proposed to investigate two novel fusion proteins aimed at overcoming complement inhibition of breast cancer cells. The recombinant fusion proteins have been successfully constructed, expressed and purified. The MUC1 vaccination study was completed, showing that C3d does function as a molecular adjuvant. Linking C3d to the tumor antigen MUC1 increased the humoral immune response and elicited a cellular immune response to the tumor. However, the T cell response was not MUC1 specific. CR2Fc has been characterized in vitro showing that it binds to C3 opsonized cells, increases complement activation and tumor cell lysis. CR2Fc has also preliminarily been characterized in vivo showing potential in a therapeutic study. Further experiments will be carried out to show the fusion proteins therapeutic efficacy and ability to modulate the immune response.

Immunotherapy Of Disease written by T.J. Hamblin and has been published by Springer Science & Business Media this book supported file pdf, txt, epub, kindle and other format this book has been release on 2012-12-06 with Medical categories.

Immunotherapy began in 1774 when the Dorset farmer Benjamin Jesty inoculated his wife and two sons with the pus from the teat of a cow suffering from cow pox, using his wife's knitting needle as a vaccinating implement. It has made slow progress. Meanwhile the science of Immunology has burgeoned so much that if all immunologists read every page of the Journal of Immunology, let alone the other Immunology journals, then they would have no time left to write for it. I am pleased that some of them have found the time to write for this volume. In spite of the rapid expansion in immuno logical knowledge and the undreamt of complexity of the immune system that has been unravelled, immunologists have remained until recently erudite but therapeutically effete. Indeed anyone purporting to treat disease by immuno logical methods has been in danger of being labelled a quack or a crackpot. Happily things are changing. The nine chapters of this volume detail nine quite different approaches to manipulating the immune system for therapeutic benefit. All are experimental and they have been attended with greater or lesser degrees of success. In some cases their main effect has been to elucidate the complexity of the problem. On the other hand, there are people alive and well today as a result of these approaches who would otherwise have perished. Immunotherapy is here to stay and it can only get better.

Immunology is a rapidly developing subject. The contributors in this book present some components of immunological knowledge which relate to the present and the possible future practice of surgery. Aspects of cellular and humoral immunity necessary for understanding are described and considera tion given to the mechanisms underlying immunological diseases. Whilst the original interest in immune response was related to microbial resistance, the recent impetus to surgical immunology has been the resurgence of organ grafting. Separate sections of the book deal with clinical organ transplanta tion, the allograft reaction, graft rejection and immunogenetics. A pre-requi site of successful organ grafting is suppressive control of immune responses. Approaches to conventional immunosuppression and specific non-reactivity are therefore considered. One method of cancer therapy is strengthening of the host's immune responses. Both experimental and clinical immunotherapy are discussed and components of tumour immunology necessary for their rational understand ing are dealt with separately. Successful immunotherapy requires monitoring of immune responses but another method for improving results of cancer treatment is earlier diagnosis by immunological methods. The book is aimed at practising surgeons who want to know the relevance of immunology to clinical surgery and laboratory scientists needing to understand the applications of their basic concepts. I am very grateful both to the authors for their contributions and the editorial staff of MTP Press Limited, for their considerable help.

Immune Aspects Of Biopharmaceuticals And Nanomedicines written by Raj Bawa and has been published by CRC Press this book supported file pdf, txt, epub, kindle and other format this book has been release on 2019-01-10 with Medical categories.

The enormous advances in the immunologic aspects of biotherapeutics and nanomedicines in the past two decades has necessitated an authoritative and comprehensive reference source that can be relied upon by immunologists, biomedical researchers, clinicians, pharmaceutical companies, regulators, venture capitalists, and policy makers alike. This text provides a thorough understanding of immunology, therapeutic potential, clinical applications, adverse reactions, and approaches to overcoming immunotoxicity of biotherapeutics and nanomedicines. It also tackles critical, yet often overlooked topics such as immune aspects of nano-bio interactions, current FDA regulatory guidances, complement activation-related pseudoallergy (CARPA), advances in nanovaccines, and immunogenicity testing of protein therapeutics.
