Coumarins are widely distributed in nature and can be found in a large number of naturally occurring and synthetic bioactive molecules. Due to the worldwide epidemic of acquired immunodeficiency syndrome (AIDS), the past ten years have witnessed a flurry of activity in the chemotherapy of viral diseases. Unprecedented scientific efforts have been made by scientists and clinicians to combat infections of human immunodeficiency virus (HIY), the causative agent. Looking back over the past ten years, we have made remarkable progress toward the treatment of the viral disease: isolation of HIV only two years after the identification of the disease, plus major strides in the areas of the molecular biology and virology of the retrovirus, etc. More remarkably, the discovery of the chemotherapeutic agent AZT (Retrovir) was made within two years after the isolation and identification of the virus, followed by unprecedented drug development efforts to culminate in the FDA approval of AZT in twenty-three months, which was a record-breaking time for approval of any drug for a major disease. The last six to seven years have particularly been an exciting and productive period for nucleoside chemists. Since the activity of AZI' was established in 1985, nucleoside chemists have had golden opportunities to discover additional anti-HIV nucleosipes, which are hoped to be less toxic and more effective than AZT, and the opportunity continues. As we all are aware, AZT possesses extremely potent anti-HIY activity, and no other nucleoside or non nucleoside has surpassed the potency of AZT in vitro.

Proteins represent one of the most abundant classes of biological macromolecules and play crucial roles in a vast array of physiological and pathological processes. The knowledge of the 3D structure of a protein, as well as the possible conformational transitions occurring upon interaction with diverse ligands, are essential to fully comprehend its biological function. In addition to globular, well-folded proteins, over the past few years, intrinsically disordered proteins (IDPs) have received a lot of attention. IDPs are usually aggregationprone and may form toxic amyloid fibers and oligomers associated with several human pathologies. Peptides are smaller in size than proteins but similarly represent key elements of cells. A few peptides are able to work as tumor markers and find applications in the diagnostic and therapeutic fields. The conformational analysis of bioactive peptides is important to design novel potential drugs acting as selective modulators of specific receptors or enzymes. Nevertheless, synthetic peptides reproducing different protein fragments have frequently been implemented as model systems in folding studies relying on structural investigations in water and/or other environments. This book contains contributions (seven original research articles and five reviews published in the journal Molecules) on the above-described topics and, in detail, it includes structural studies on globular folded proteins, IDPs and bioactive peptides. The progress of society has led to an improvement of the quality of life of a significant number of people. The book starts with the editors' preface summarizing the contributions of each of the following thirteen articles dealing with chemical and pharmacological aspects of the molecular modulators of the oxidative stress in regard to various therapeutic approaches in cardiovascular and neurodegenerative diseases, cancer, and diabetes.
