Hydrogels have been widely used as 3D niche for cell delivery and tissue engineering. However, most hydrogels are nanoporous, whereas macroporosity is desirable for efficient nutrient diffusion, cell proliferation and matrix deposition. Though porogens may be used to introduce macroporosity, they further decrease the already weak mechanical properties of the hydrogels. To overcome these limitations, we have recently reported a method to fabricate gelatin hydrogels into microribbon (u00b5RB)-like structures using wet-spinning, followed by further photocrosslinking to form a macroporous scaffold. We have demonstrated that such 2-step fabrication process allows homogeneous encapsulation of stem cells inside 3D macroporous scaffolds and the unique u00b5RB morphology confers markedly enhanced mechanical strength to the scaffolds. While our previous work demonstrates the advantage of macroporosity in hydrogels for enhancing stem cell-based tissue regeneration using gelatin u00b5RBs, previous method was not suitable for fabricating other ECM polymers into u00b5RB structures, and how tuning u00b5RB compositions impact MSC-based bone formation in 3D remains unknown. The goals of this study are to develop a method that enable fabricating various extracellular-matrix (ECM) compositions into u00b5RB-like structures, and assess the effect of ECM composition on osteogenesis of mesenchymal stem cells (MSCs) and bon matrix production for bone repair. We demonstrated successful syntheses of u00b5RBs from various ECM compositions for forming macroporous scaffolds. Gelatin u00b5RB alone lead to high collagen, low mineralization; while GAG u00b5RB alone lead to low collagen, high mineralization. Co-spun u00b5RBs, but not mixed gelatin and GAG u00b5RBs, led to synergized bone formation with robust mineralization and collagen deposition. In summary, u00b5RB-based scaffolds with tunable compositions provide novel injectable macroporous matrices to enhance stem cell based bone regeneration.

Cartilage injuries in children and adolescents are increasingly observed, with roughly 20% of knee injuries in adolescents requiring surgery.
