Although post-structuralism has highlighted the importance of what is offstage, lost, forgotten, hidden or discarded, silent or silenced, the poetics and politics of absence (much like its ethics and aesthetics) have rarely been discussed across media or disciplines. The book conceptualizes 'radical absence' to describe a certain tradition of resistance to ontology, predication, and representation, contesting their reliance on a metaphysics of presence. Apophatic speech, empty signifiers, and figural voids are some of the figures through which radical absence becomes apparent, with unprecedented intensity, in 20th-century theory, literature, film, and the arts. Phantasmatic and outrageous, such figures play with creative strategies of de-materialization, irony, and other forms of discursive undoing. Therefore, absence becomes more than a simple theme; it reflects back on the medium and the meaning-making conditions under which it operates. Elusive and imprecise as an object of study, absence is in need of more subtle and flexible epistemological frameworks. The author proposes to think it not only as a counter-concept for presence, but also - perhaps more productively - as infinite spacing, deferral, fragmentation, and displacement.

Orientalism and the Figure of the Jew proposes a new way of understanding modern Orientalism. Tracing a path of modern Orientalist thought in German across crucial writings from the late eighteenth to the mid–twentieth centuries, Librett argues that Orientalism and anti-Judaism are inextricably entangled. Librett suggests, further, that the Western assertion of “material” power, in terms of which Orientalism is often read, is overdetermined by a “spiritual” weakness: an anxiety about the absence of absolute foundations and values that coincides with Western modernity itself. The modern West, he shows, posits an Oriental origin as a fetish to fill the absent place of lacking foundations. This fetish is appropriated as Western through a quasi-secularized application of Christian typology. Further, the Western appropriation of the “good” Orient always leaves behind the remainder of the “bad,” inassimilable Orient. The book traces variations on this theme through historicist and idealist texts of the nineteenth century and then shows how high modernists like Buber, Kafka, Mann, and Freud place this historicist narrative in question. The book concludes with the outlines of a cultural historiography that would distance itself from the metaphysics of historicism, confronting instead its underlying anxieties.

The remarkable ability to target one or even a couple of sites on a large protein with a small molecule, under photochemical control, is a considerable challenge and this challenge has been addressed in some depth in this book. Systematic modification of the structure of the photoreagent provided valuable information on the binding site recognition as well as the mechanism of the photocleavage reaction. Some factors that impact the photocleavage reaction include the exact location of the probe binding site on the protein, conformations of the bound probe at the binding site, protein size, functional groups present on the probe that interact with the protein surroundings either in a favorable or unfavorable manner, overall charge on the photoreagent, and photophysical as well as photochemical properties of the probe. The protein photocleavage studies, in all case, were preceded by detailed binding studies by a variety of spectroscopic methods. Methods as simple as absorption and fluorescence spectroscopies or more sophisticated circular dichroism spectroscopy were used. Conclusions that are most consistent with the binding data indicated a single binding site on most proteins, irrespective of the probe or the protein, with only one exception noted so far. Photoactivation of the bound probe resulted in protein cleavage at a single site, in many instances. The specificity for the reaction has been investigated in detail and molecular modeling studies provided a firm ground to rationalize the observed cleavage sites. The reagents provide unique tools for sequencing large proteins by converting them into smaller fragments by non-biochemical transformations. Understanding of the rules for the above methodology are also investigated which provided rational methods for the design of small molecules that could bind at particular sites on large proteins, and this is a major breakthrough for a variety of fields including drug design, protein targeting, mass spectrometry, proteomics and other cutting-edge research areas.
