Cells have evolved multiple strategies to adapt the composition and quality of their protein equipment to needs imposed by changes in intra- and extracellular conditions. The appearance of pro teins transmit ting novel functional properties to cells can be controlled at a transcrip tional, posttranscriptional, translational or posttranslational level. Extensive research over the past 15 years has shown that transcriptional regulation is used as the predominant strategy to control the production of new proteins in response to extracellular stimuli. At the level of gene transcription, the initiation ofmRNA synthesis is used most frequently to govern gene expression. The key elements controlling transcription initiation in eukaryotes are activator proteins (transactivators) that bind in a sequence-specific manner to short DNA sequences in the of genes. The activator binding sites are elements of larger proximity control units, ca lied promoters and enhancers, which bind many distinct proteins. These may synergize or negatively cooperate with the activators. The do novo binding of an activator to DNA or, if already bound to DNA, its functional activation is what ultimately turns on a high-level expression of genes. The activity of transactivators is controlled by signalling pathways and, in some cases, transactivators actively partici pate in signal transduction by moving from the cytoplasm into the nuc1eus. In this first volume of Inducible Gene Expression, leading scientists in the field review six eukaryotic transactivators that allow cells to respond to various extracellular stimuli by the expression of new proteins.

This is the first of two volumes which aim to cover all established eukaryotic transcription factor systems that are direct targets for the signal transduction pathways. Leading molecular biologists contribute reviews on topics which take a broad view, and which should be of interest to students and advanced researchers in biology and medicine. Chapters attempt to answer such fundamental questions as: what is the signal and how and when is it produced? What is the receptor, and what further signalling molecules are involved? What is the biochemistry and molecular biology of the transcription factor that is the ultimate target of the signalling pathway? What is the physiological role of factor?

Over the past few years there have been considerable advances in our understanding of cellular control mechanisms, and current research is now linking areas of biology that were previously thought of as being quite separate. Molecular Aspects of Cellular Regulation is a series of occasional books on multidisciplinary topics which illustrate general principles of cellular regulation. Previous volumes described Recently Discovered Systems of Enzyme Regulation by Reversible Phosphorylation (Volumes 1 and 3), The Molecular Actions of Toxins and Viruses (Volume 2), Molecular Mechanisms of Transmembrane Signalling (Volume 4) and Calmodulin (Volume 5). This sixth volume, The Hormonal Control of Gene Transcription, has now been published to highlight recent important advances in our understanding of this topic which is linking two of the most active areas of current biochemical and molecular biological research (hormone action and gene transcription) and leading to the emergence of unifying concepts.

This new volume on gene expression and epigenetics discusses environmental effects related to specific gene expression. The book also shows methods for bioinformatic analysis of the epigenome. The book is broken into two sections: the first looks at eukaryotic DNA methylation and the second addresses how to integrate genomic medicine into clinical practice. The book includes chapters on these topics: • Gene expression in colon cancer tissue • Epigenetics in human acute kidney injury • Embryologically relevant candidate genes in MRKH patients • DNA methylation in common skeletal disorders • Causal relationships in genomics • Predicting severe asthma exacerbations in children • Epigenetic understanding of gene-environment interactions in psychiatric disorders

For as much as we know about DNA and gene expression, many more mysteries remain to be solved. Epigenetics and epigenomics seek to study heritable modifications in gene expression that do not involve underlying DNA sequences to further human health changes. Examining the Causal Relationship Between Genes, Epigenetics, and Human Health provides innovative research methods and applications of chemical activation or deactivation of genes without altering the original DNA sequence. While highlighting topics including gene expression, personalized medicine, and public policy, this book is ideal for researchers, geneticists, biologists, medical professionals, students, and academics seeking current research on the expanding fields of genomics, epigenomics, proteomics, pharmacogenomics, and genome-wide association studies.

The field of neuroendocrinology has extended from the initial interest in the hypothalamic control of pituitary secretion to embrace multiple reciprocal interactions between the central nervous system and endocrine systems in the coordination of homeostasis and various physiological responses from adaptation to disease. Most recently, epigenetic mechanisms were recognized for their role in the development of the neuroendocrine axes as well as in the mediation of gene-environment interactions in stress-related psychiatry disorders.
