Fungi of the order Pucciniales cause rust diseases on many plants including important crops and trees widely used in Agriculture, forestry and bioenergy programs; these encompass gymnosperms and angiosperms, monocots and dicots, perennial and annual plant species. These fungi are obligate biotrophs and -except for a few cases- cannot be cultivated outside their hosts in a laboratory. For this reason, standard functional and molecular genetic approaches to study these pathogens are very challenging and the means to study their biology, i.e. how they infect, develop and reproduce on plant hosts, are rather limited, even though they rank among the most devastating pathogens. Among fungal plant pathogens, rust fungi display the most complex lifecycles for with up to five different spore forms and for many rust fungi, alternate hosts on which sexual and clonal reproduction are achieved. The genomics revolution and particularly the application of new generation sequencing technologies have greatly changed the way we now address biological studies and has in particular accelerated and made feasible, molecular studies on non-model species, such as rust fungi. The goal of this research topic is to gather articles that present recent advances in the understanding of rust fungus biology, their complex lifecycles and obligate biotrophic interactions with their hosts, through the means of genomics. This includes genome sequencing and/or resequencing of isolates, RNA-Seq or large-scale transcriptome analyses, genome-scale detailed annotation of gene families, conformation of genes and/or expression of gene complements via proteomics, and comparative analyses among the various rust fungi and, where feasible, with other obligate biotrophs or fungi displaying distinct trophic modes. This Research Topic provides a great opportunity to provide an up-to-date account of rust fungus biology through the lens of genomics, including state-of-the-art technologies developed to achieve this knowledge.

This book describes approaches for the analysis of plant pathogenic fungi that range from ecosystem composition to in vitro and wet-lab to computational analyses. Chapters detail bioinformatics protocols for genome assembly, transcriptome analysis and small RNA profiling, characterization of secondary metabolome, analysis of post-translational modification and localization of proteins, examination of virulence function and interaction with plants or other microorganisms, and targeted mutagenesis using various approaches. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Authoritative and cutting-edge, Plant Pathogenic Fungi and Oomycetes: Methods and Protocols aims to provide techniques developed through a wide range of perspectives to facilitate research on a comprehensive understanding of these important filamentous pathogens.

Genomics Of Plant Associated Bacteria written by Dennis C. Gross and has been published by Springer this book supported file pdf, txt, epub, kindle and other format this book has been release on 2014-06-16 with Science categories.

This book describes how genomics has revolutionized our scientific understanding of agriculturally important plant-associated bacteria. Each chapter focuses on the genomics of particular bacteria: the first described plant pathogen, Erwinia amylovora; phytoplasmas lacking cell walls; fastidious, phloem-restricted liberibacters; Pseudomonas syringae, which is a genetically tractable model system; Xanthomonas citri, which causes a disease that can devastate citrus crops and Pseudomonas fluorescens, which can protect plants from diseases. Topics considered in this volume include the importance of horizontal gene transfer in originating new bacterial strains and species and advances in transcriptomics that allow us to describe the complex regulatory networks critical to plant-microbe interactions. The availability of the Xanthomonas oryzae genome has led to new technologies in genome editing, which will revolutionize approaches to genetic engineering, even in eukaryotes. The contributions show how genomics has greatly accelerated progress toward understanding the biology of these bacteria and how that understanding can be translated into novel crop protection methods.

This book is specially written for researchers at various levels, for example, in forestry, agriculture, industry, university and college laboratories. It describes the fungal pathogenicity; resistance behavior of fungal biofilms and its mechanisms; different categories of fungal infection and colonization patterns with example relevant to soybean; characteristics of white rob of corncob and head smut of maize such as cycle, pathogenicity factors, control methods, the abilities of chitosan and its derivatives to elicit resistance reactions in plants and its action in the production and viability of fungal spores; and the mode of actions of single constituents of different essential oils depending on different case studies. In addition, this book also describes the importance of synthetic peptides as an alternative tool for the diagnosis of cryptococcosis. Finally, a survey of fungal diseases occurring on trees of Namibia is described. This survey is the first dedicated step to find ways of protecting them from disease-causing agents.
