This book presents an investigation of gigaseal formation using micro/nanotechnology. The aims of the book are twofold. First, it explains the mechanisms of gigaseal formation using the latest discoveries. Second, it provides practical techniques for frequent formation of high resistance seals. The formation of a high-resistance electrical seal, also known as a gigaseal, between a cell membrane and a glass micropipette tip is essential in patch-clamp experiments. Even though four decades have passed since the introduction of the patch-clamping technique by Neher and Sakmann, gigaseal formation remains an obstacle in developing the high-throughput ion channel screening systems required by the pharmaceutical industry. Here the authors share their latest methods for achieving gigaseal formation and describe techniques that are highly desirable at both research and industrial levels. Ion channel research has increased tremendously in the past 35 years since the first publication of the patch clamp technique by Neher and Sakmann in 1976. This is documented by the rising number of publications listed in Pubmed (http://www.ncbi.nlm.nih.gov/pubmed) including the keyword ‘ion channel’ from just 186 hits in 1976 to almost 180,000 hits today. Ion channels attract this great interest due to their pivotal role in the control of fundamental physiological processes in a plethora of different tissues. Moreover, their importance in a wide range of inherited and drug-induced pathologies spanning all major therapeutic areas makes them attractive targets for pharmacological drug screening and potential risk factors when assessing drug safety (Ashcroft, 2006; Clare, 2010; Dunlop 2008; Milligan 2009). Several methods and technologies have been developed to meet the analytical needs for studying ion channels. These approaches have addressed ion channel function directly as well as in the context of the cell and tissue. Scaling of these technologies has allowed ion channel analysis to be carried out on high throughput and high content assay systems. Individual cells behave in surpnsmg ways that cannot be deduced from the averaged results of an organ as assessed by the use of conventional biochemical methods. Thus multicellular plant and animals systems are being investigated by an increasing array of histochemical and cytochemical techniques based on general chemical or specific immunological interactions to identify structural materials and to assess biological activities. In recent years there has been an increasing range of fluorescent probes, along with advanced computerised imaging and analysis techniques, which allows the behaviour of individual living cells to be followed in considerable detail. The parallel use of microinjection, microelectrodes and patch-clamping provides additional information about cells and their responses. Recombinant DNA technology has highlighted the desirability and the power of microinjecting defined materials into specific cells and so manipulating their fundamental biochemistry. New hypotheses are being tested which will form the cornerstone of future developments across the whole spectrum of biotechnology. The First European Workshop on Biotechnology Applications of Microinjection, Microscopic Imaging and Fluorescence was run at the University of East London, U.K, 21st-24th April, 1992 with the objective of bringing together a diverse group of individuals who were using these state-of-the-art applications for biotechnological exploration. A novel feature of the meeting was paiticipation by instrument manufacturers in the programme: there were hands-on workshops (where living cells could be examined), combined with the poster sessions.
