Glycobiology has its roots in the nineteenth century, when chemists first began to analyze sugar and polysaccharides. Advances in this area continued at a steady rate during most of this century, but the past 20 years has witnessed an unparalleled explosion of new knowledge that has transformed the field. This monograph contains the basic information needed to understand the field of glycobiology along with the most current work at the forefront of the field.

A Glance at Currently Accepted “Hallmarks of glycosylation in cancer & Future Perspectives”. The hallmarks of cancer were originally outlined in 2000 and comprised six biological capabilities acquired during the multi-step development of cancer that allow cancer cells to: · Survive, · Proliferate and · Ddisseminate. Aberrant glycosylation in cancer was first described more than 45 years ago, and since then it has been well documented that fundamental changes in the glycosylation patterns of cell surface and secreted glycoproteins occur during malignant transformation and cancer progression. Glycans have roles in cancer cell signalling, tumour cell dissociation and invasion, cell-matrix interactions, angiogenesis, metastasis and immune modulation. Aberrant glycosylation is often cited as a ‘hallmark of cancer’ but is notably absent from both the original hallmarks of cancer and from the next generation of emerging hallmarks. Thus, it is strived here in this Booklet to discuss how glycosylation is clearly an enabling characteristic that is causally associated with the acquisition of all the hallmark capabilities. …Dr. H. This book, now in an extensively revised second edition, provides a comprehensive summary of the latest knowledge regarding glycosignals and a thorough analysis of their involvement in not only cancers but also other refractory conditions such as chronic inflammatory disorders. Many relevant topics are covered, including the search for novel tumor epitopes related to carbohydrates, the assembly of glycoconjugates, the modulation of signaling pathways by glycosylation, and interactions between complex carbohydrates and their recognition molecules. The role of various research approaches, for example advanced mass spectrometry, high-resolution imaging, and bioinformatics, is closely examined, and the results of novel therapeutic trials targeting glycosignals are discussed. The book will be essential reading for students and young researchers with an interest in glycoscience. In presenting new results and approaches and identifying areas for future research, it will also be of benefit for specialists in the field.

Changes in glycosylation are a universal characteristic of malignant transformation, and tumor-specific glycan structures (which we denominate as the tumor glyco-code) are strongly associated with cancer progression and immune-modulation. Because immune cells express a large variety of glycan-binding receptors (called lectins), they can sense and respond to changes in the glycan signature of their environment; which often leads to the induction of inhibitory immune processes in those cells (Chapter I). In this thesis, we focus in the study of the tumor glyco-code of pancreatic ductal adenocarcinoma (PDAC) that represents one of the most aggressive malignancies, with a 5-year survival rate of only 9%. By analyzing publicly available transcriptomic data of patient samples, we studied two specific glycosylation signatures that correlate with progression, clinical outcome, and the modulation of the immune system: Fucosylation (Chapter II) and Sialylation (Chapter IV). In Chapter II, we identified that the presence of fucose-containing structures in cancer cells are associated with differences in their epithelial-to-mesenchymal transition status, the clinical outcome of patients and previously published molecular subtypes of PDAC. Moreover, these fucosylated structures can serve as ligands for the receptor DC-SIGN, present in tumour-associated macrophages (TAMs), and modulate their activation by inducing the overexpression of the anti-inflammatory cytokine IL-10. On the other hand, one of the glycan moieties generally overexpressed in cancer are sialic acids, which can modulate the function of immune cells via interaction with Siglec receptors (Chapter III).

Diverse alterations of glycosylation occur in diseases such as cancer, metastasis, leukemia, inflammatory and other diseases. The glycosylation abnormalities found in disease are the result of complex rearrangements of the oligosaccharide assembly by glycosyltransferases. This volume reviews several mechanisms that may underly the extremely complex alterations in disease. Disease specific glycosylation may contribute to the disease process by altering cellular functions, or may be exploited therapeutically. Specific therapy may be aimed at correcting glycosylation abnormalities based on knowledge of the mechanisms leading to the disease phenotype and the three-dimensional interactions between carbohydrates and carbohydrate-binding molecules.
