GTPases that regulate the myriad of membrane fission events that facilitate the assembly and disassembly of COPII (Sar1), COPI (ARF) and clathrin coats (dynamin) involved in exocytic and endocytic trafficking pathways and mitochrondrial organization remain a major challenge for future investigations to understand membrane architecture of eukaryotic cells. This volume of MIE provides a comprehensive set of articles describing the use and application of state-of-the-art methodologies to identify and characterize these GTPases and their rapidly expanding list of regulators and effectors. Methodologies focused on biochemical, molecular and advanced imaging techniques provide a wealth of investigational tools for those currently in the field and those entering the field. Many of the methodologies are generally applicable to study of these GTPases in vitro and in vivo to elucidate function in regulation of cell proliferation and signaling in normal tissue and in disease. *Comprehensive collection of GTPases (ARF, Sar1, Dynamin) GTPases involved in vesicle coat assembly and membrane fission *Identification and characterization of GTPase GEFs, GAPs and effectors *General methodologies to study GTPase function in vitro and in vivo

Rab GTPases now comprise a family of >63 members. They are emerging as the key hub element controlling the membrane architecture of eukaryotic cells. They are intimately involved in vesicle targeting and fusion in both the endocytic and exocytic pathways and direct the assembly and disassembly of protein complexes that include regulators (GEFs and GAPs), effectors (tethers/motors) and fusion components (SNAREs) that control membrane targeting and fusion. During the last 3 years the field has virtually exploded with the identification and characterization of many new Rab proteins and their effectors. Our understanding of how Rab GTPases control membrane function remains at its infancy. This volume of Methods in Enzymology, GTPases Regulating Membrane Targeting and Fusion, provides a wealth of new concepts, approaches and tools to study Rab proteins in the test tube and in living cells that will be of strong benefit to both established laboratories and new investigators in the field to elucidate Rab GTPase function in cellular development, differentiation and proliferation. Comprehensive overview of Rab GTPase phylogeny and systems biology Identification and characterization of Rab GEFs, GAPs and effectors General methodologies to study Rab GTPase function in vitro and in vivo using biochemical, molecular and microscopy approaches

Pathogenic bacteria for human and animals have developed sophisticated weapons, termed virulence factors, to ensure their replication and persistence into their hosts. The authors in this volume show a synthesis on how the various host cellular Rho GTPases activities are manipulated by bacteria to fulfil their virulence.

For the first time experts in the area of signalling research with a focus on the ARF family have contributed to the production of a title devoted to ARF biology. A comprehensive phylogenetic analysis of the ARF family, tables of the ARF GEFs and ARF GAPs, and more than a dozen chapters describing them in detail are provided. The impact of the ARF proteins on widely diverse aspects of cell biology and cell signalling can be clearly seen from the activities described; including membrane traffic, lipid metabolism, receptor desensitization, mouse development, microtubule dynamics, and bacterial pathogenesis. Anyone interested in understanding the complexities of cell signalling and the integration of signalling networks will benefit from this volume.

Membrane Dynamics And Domains written by Peter J. Quinn and has been published by Springer Science & Business Media this book supported file pdf, txt, epub, kindle and other format this book has been release on 2013-06-29 with Science categories.

The fluid-mosaic model of membrane structure formulated by Singer and Nicolson in the early 1970s has proven to be a durable concept in terms of the principles governing the organization of the constituent lipids and proteins. During the past 30 or so years a great deal of information has accumulated on the composition of various cell membranes and how this is related to the dif ferent functions that membranes perform. Nevertheless, the task of explaining particular functions at the molecular level has been hampered by lack of struc tural detail at the atomic level. The reason for this is primarily the difficulty of crystallizing membrane proteins which require strategies that differ from those used to crystallize soluble proteins. The unique exception is bacteriorhodopsin of the purple membrane of Halobacterium halobium which is interpolated into a membrane that is neither fluid nor in a mosaic configuration. To date only 50 or so membrane proteins have been characterised to atomic resolution by diffraction methods, in contrast to the vast data accumulated on soluble proteins. Another factor that has been difficult to explain is the reason why the lipid compliment of membranes is often extremely complex. Many hundreds of different molecular species of lipid can be identified in some membranes. Remarkably, the particular composition of each membrane appears to be main tained within relatively narrow limits and its identity distinguished from other morphologically-distinct membranes.
