In the summer of 1988, my developmental biology professor announced to the class that hematopoietic stem cells (HSCs) had finally been purified. Somehow, I never forgot the professor’s words. When I started working in Dr. Irv Weissman’s labo- tory at Stanford as a postdoctoral fellow, I realized that the findings mentioned by the professor were from Weissman’s laboratory and had been published in a 1988 edition of the journal Science. It has been over 20 years since the publication of that seminal paper, and since then tremendous advances in understanding the biology and maturation of HSCs, namely the process of hematopoiesis, which includes lymphocyte development, have been made. These discoveries were made possible in part by advancements in technology. For example, recent availability of user friendly fluorescence activated cell sorting (FACS) machines and monoclonal an- bodies with a variety of fluorescent labels has allowed more scientists to sort and analyze rare populations in the bone marrow, such as HSCs. All classes of hematopoietic cells are derived from HSCs. Stem cell biology draws enormous attention not only from scientists, but also from ordinary people because of the tremendous potential for development of new therapeutic application to diseases that currently lack any type of effective therapy. Thus, this type of “regenerative medicine” is a relatively new and attractive field in both basic science and clinical medicine.

This book renders a comprehensive understanding of hematopoietic stem cells (HSCs) from their embryonic development through adult maintenance to aging, in the studies conducted in zebrafish and mammals. Hematopoiesis provides a paradigm for understanding the development, maintenance, regeneration, aging and malignant transformation of mammalian organs. Sitting at the apex of the hematopoiesis hierarchy tree, HSCs orchestrate their proliferation, self-renewal, and differentiation to produce all the blood cell lineages throughout life, which represents the best example for somatic stem cell studies. In this book, key regulatory mechanisms for HSC self-renewal and differentiation are overviewed in an array of fields including epigenetics, metabolism and microenvironment regulation. It also highlights the HSC heterogeneity and clonal dynamics from the recent advanced single-cell technologies. This book elaborates on the research history of HSC studies and reveals how the insights from HSC studies shed light on their clinic application. It presents great value from the bench to the clinic.

Blood has long been viewed as a conduit for therapy, stemming from the ancient days of phlebotomy to remove evil humors to the development of successful blood transfusions to replace missing blood components. The identification and characterization of hematopoietic stem cells by Drs. Till and McCulloch revolutionized the field and soon after, non-hematopoietic stem and progenitor cells were characterized from the blood and bone marrow. Some of these cell types and various blood-derived cell lineages are involved in the repair of various types of tissue damage that span the spectrum of medical disorders. The goal of this book is to provide an up-to-date review of the various types of blood-derived cells with regenerative capacity, identify opportunities for intervention by examining specific clinical applications, and recognize the regulatory environment that will encompass future therapies in regenerative medicine.

The dogmas of the quiet past are inadequate to the stormy present. The occasion is piled high with difficulty, and we must rise with the occasion. As our case is new, so we must think anew and act anew. Abraham Lincoln, 1862 When I came across this quote, it made me recall my first participation at an international meeting on bone marrow transplantation, at a time when this was the only term that was used to describe the field. During a particular session there was a presentation on the use of peripheral blood as the sole source of stem cells for transplantation, and a member of the audience rose to state that it was medically unethical to consider such treatment, as it certainly could not contain stem cells. Now nearly twenty years later, peripheral blood is the predominant source of stem cells used for hematopoietic stem cell transplantation. In the same period of time there have been several other dogmatic opinions, which permeate all of medicine, that have come and gone in the field of hematopoietic stem cell transplantation, and will continue to do so with advancements from basic and clinical research. It is within this context that the format of this book was devised. Traditionally reviews on specific topics related to hematopoietic stem cell transplantation reflect the views of a single author or a research group.

Although much is known with respect to blood cell formation and function, many new concepts in the areas of the regulation of hematopoietic stem cell commitment and the subsequent survival, proliferation, and differentiation of progenitors have been elucidated in the last five years. Our understanding of the microenvironment where stem cells reside and commit to distinct blood types (the niche) has grown significantly in recent years. Furthermore, blood cells have been used as the key model system to study microRNA function and the role of microRNAs in the transformation of normal cells into cancer cells. The current volume Molecular Basis of Hematopoiesis, edited by Amittha Wickrema & Barbara Kee, provides the most recent developments in the area in addition to a chapter on the utilization of basic science knowledge for the treatment of blood diseases. Each chapter in this book has been written and edited by faculty in major academic and research institutions around the world, who are pushing the frontiers of research in this important area.
