Even though diffusion chamber culture was commenced long before ortho dox tissue culture by Metchnikoff (1887) there have been only sporadic attempts to use this methodology to study cell proliferation (review by Carsten, Chap. 1). Not so long ago diffusion chamber culture was nicknamed "confusion chamber" culture. I believe this conference has removed the confusion and will truly point out the intrinsic value of the system. It is not a substitute for established in vitro culture nor for in vivo studies. It complements both. Dr. Arne B~yum introduced diffusion chamber culture at Brookhaven National Laboratory. After some modest success in showing that one could culture human bone marrow and with appropriate stimuli induce erythro poiesis in diffusion chambers, several of the participants at this conference visited Brookhaven to learn firsthand this simple technology and to apply it in their own laboratories. However, the technique did not spread widely and controversy arose in which the same question was repeatedly asked: Can the diffusion chamber technique provide information that is not ob tainable more rapidly and easily, and at less expense by the in vitro tech niques? As a result of our deep interest in and involvement with diffusion chamber culture Dr. A. L. Carsten and I organized this conference. A major objective of this conference was to seek answers to the above question.

This translation from Russian into English of the monograph Hematopoi etic and Lymphoid Tissue in Culture by E. A. Luriya provides us for the first time with a comprehensive review of the massive amount of work carried out in the USSR in this important area of biological research. Historically, the pioneering tissue culture studies of Maksimov, Popov, Chasovnikov, and their colleagues working in Russia in the early part of this century provided much of the impetus for subsequent studies on lymphopoiesis and hematopoiesis in vitro and much of the basic information used subsequently to achieve immune responses in vitro and to develop the methodology for cloning hematopoietic stem cells. Particularly welcome in this translation is the addition of the Appendix, in which Drs. Luriya and Fridenshtein describe in some detail the methodology that has been developed in their laboratories for the study of a variety of cell types in cell and tissue culture. For many of us, this description provides information that had previously not been accessible to us: porosity of millipore fIlters in relation to growth properties; colony-forming cell adhesiveness; pro liferation kinetics of stromal cells; differential effectiveness of various media used for culture of liver cells; details of washing procedures for fIlter substrates used in culture.

Hematopoiesis, or the process of blood formation, has been extensively studied at both basic and clinical levels. Human diseases such as thalassemia, immunodeficiency, and leukemia represent defects in this process. Approaches to treat these disorders have required a basic understanding of the biology of blood cells. For instance, hemapoietic stem cell replacement or bone marrow transplantation has been used to ameliorate disease. This volume focuses on hematopoiesis at a cellular and molecular level, and establishes the basis for clinical manipulation of hematopoietic cells for therapeutic benefit. In Part I, the cellular characteristics of progenitors and stem cells are explored. Emphasis is placed on purification of stem cells and both in vitro and in vivo assays. The regulation of normal and leukemis stem cells is illustrated. An excellent discussion of potential use of these cells for gene therapy concludes this section. Hemapoiesis is easily studied during embryogenesis. Part II develops the concept of the waves of hemapoiesis during development. Comparative hematology is making a major comeback as a field in the 1990's. One hope is that general principles of hematopoiesis will be established by studying many models and systems. Part III delves into critical factors that regulate hematopoiesis, including both intracellular and extracellular signals. Part IV and V describe lineage programs for myeloid and lymphoid lineages. These chapters are meant to be illustrative of the different cell fates, but are not exhaustive. Part VI examines the genetics of hematopoisis, particularly in animal models. The hematopoietic system is in constant contact with stromal cells and endothelial cells during development and in the adult. Evidence suggests that endothelial cells and blood cells may arise from a common progenitor, the hemangioblast. Part VII and VIII discuss the stromal and endothelial cells with the emphasis on their interaction with hematopoietic cells.

The daily production of hundreds of billions of blood cells through the process of hematopoiesis is a remarkable feat of human physiology. As a consequence, many pathological conditions are attributable to blood cell abnormalities, and a fair number of these are now clinically treatable as a direct result of hematopoietic research. Proliferation of hematopoietic stem cells, and their differentiation into the many different lineages of functional mature cells, is highly regulated and responsive to many environmental and physiological challenges. Our relatively advanced understanding of this stem cell system provides potentially important insights into the regulation of development in other tissues, many of which are now being acknowledged as stem ce- based, perhaps even into adulthood. The recent public and scientific fanfare following announcement of human embryonic stem cell studies suggests that stem cell research will continue to be a relevant and exciting topic.
