The traditional view of sensory processing assumes a bottom-up flow of information to higher cortical regions, to construct representations of the world. However, emerging evidence emphasizes the role of expectations, revealing the brain's active involvement in predicting sensory input beyond the traditional view. Predictive coding (PC) integrates this by utilizing generative models based on prior experiences, encoded in higher cortical areas. Mismatches between predictions and sensory evidence update these models, facilitating Bayesian optimization of beliefs about the sensory world. However, the precise neural mechanisms of predictive processing remain unclear. In this PhD thesis, I developed an audio-visual paradigm within the framework of PC to investigate these mechanisms. Firstly, employing EEG, Bayesian modeling, and machine learning, I show that alpha oscillations play a key role in generating predictions. Additionally, I demonstrate that low-dose ketamine, an NMDA-receptor blocker, disrupts behavioral indices of predictions, their representation in frontal cortex, and pre-activation of sensory templates in higher-order sensory cortex. Secondly, I explore how predictions are communicated to higher-order sensory cortex and consequences of a prediction-sensory evidence mismatch. Utilizing electrocorticographic recordings in medically refractory epileptic patients, I show the transmission of predictions through traveling alpha waves from higher-to-lower-order cortices. Moreover, the occurrence of a mismatch enhanced inter-trial phase coherence, increased high-gamma power, and induced cross-frequency coupling in sensory cortex. Additionally, alpha oscillatory waves propagated from lower-to-higher-order cortices, to update existing models. Thirdly, I investigate how the brain acquires accurate internal models of the world, elucidating the progression from initial weak and inaccurate models to precise models. Using a dynamical systems approach, I show that learning about priors induced changes in neural state space trajectories, where the distance traveled in this space correlates with the predictive information's strength. Finally, I propose a theoretical framework in which the cerebellum acts as a nudged particle filter to generate internal models of the world. This framework provides insight into how the brain integrates diverse experiences to form a causal model of our surroundings. In conclusion, this thesis sheds light on the neural mechanisms underpinning predictive processing in humans, contributing to our understanding of predictive processing's role in perception and decision-making.
