Hormones, Brain Function, and Behavior brings together the proceedings of a conference on neuroendocrinology held in May 1956 in New York. The papers explore selected aspects of hormone actions in relation to brain function and behavior and cover topics ranging from the effects of steroid hormones on the nervous system to the behavioral consequences of sex hormones and thyroid hormones. Serotonin, epinephrine, and their metabolites are also discussed in relation to experimental psychiatry. This book brings together some of the results and ideas produced by a large number of people-colleagues and students with whom I am privileged to work in the laboratory at Rockefeller University. In terms of my personal history I see it as a confluence of creative forces persons from whom I have learned. I was instructed in neuroanatomy by Walle J. H. Nauta at M. I. T. , and later in a course at Harvard Medical School under the direction of Richard Sidman. At Harvard Medical School, where M. I. T. graduate students were allowed to cross register, the superb neurophysiology course was under the guiding spirit of Stephen Kuffler. Later, I benefited greatly from participating in his summer course in electrophysiological techniques at Woods Hole. Eric Kandel and his colleagues have provided us with the most exciting contemporary approach to the conceptualization and study of cellular mechanisms for behavior. Here at Rockefeller, Carl Pfaffmann and Neal Miller have been leaders in every sense of the word. Not only did they provide me with opportunities to grow to scientific maturity; they also set an example of clear thinking about mechanisms for mammalian behavior patterns. I wrote this book to show how the systematic use of increasingly detailed electrophysiological, neuroanatomical, and neuroendocrine tech niques can explain the mechanism for a mammalian behavioral response. The behavior in question happens to be sensitive to steroid hormones and plays a central role in reproduction.

It is well known that the class of steroid hormones known as estrogens have powerful effects on organs related to reproduction such as the uterus and the breast. What is less well known is that estrogens also profoundly modulate brain function and behavior. Estrogens, such as estradiol, can occur in brain as the result of ovarian secretion of the hormone into the blood that then finds its way to the brain. In male vertebrates, the testes secrete androgens, such as testosterone, into the blood and this class of steroid hormones can be converted into estrogens in the brain via the action of the enzyme aromatase which is expressed in the male brain in many species. Finally estradiol can be synthesized de novo from cholesterol as it has been shown in a variety of species that all the enzymes required to synthesize estrogens are expressed in the brain. This book collects chapters by experts in the field that considers, how estradiol is synthesized in the brain and what its effects are on a variety of behaviors. Special attention is paid to the enzyme aromatase that is distributed in discrete regions of the brain and is highly regulated in a sex specific and seasonal specific manner. Recently it has become clear that estrogens can act in the brain at two very different time scales, one is rather long lasting (days to weeks) and involves the modulation of gene transcription by the hormone-receptor complex. A second mode of action is much quicker and involves the action of estrogens on cell membranes that can result in effects on second messenger systems and ultimately behavior within minutes. Thus this book highlights novel views of estrogen action that are still under-appreciated namely that estrogens have significant effects on the male brain and that they can act on two very different times scales. This volume will be of interest to both basic researchers and clinicians interested in the action of estrogens.

Peptide hormones are involved in mental performance. pain perception, food and water metabolism. sleep. sexual behaviour and nursing behaviour, and disturbances in the hormonal climate of the brain may be associated with psychopathology. cognitive disturbances and. possibly. addic tion. The clinical influence of neurohypophysial hormones and their frag ments is studied today on learning and memory. on inadequate behaviour and addiction, in Parkinson's disease. Lesch-Nyhan syndrome. depression and schizophrenia. Fragments of adrenocorticotrophin have been shown to affect motivation, concentration and attention. and neuropeptides derived from ~-lipotrophin are probably involved in psychopathology. Thyrotrophin releasing hormone has been implicated in depression. and lutein-releasing hormone in sexual disturbances. In spite of the impressive experimental data; clinical results to date have been controversial and. to some extent. anecdotal. In some cases they have been exciting. and in others disappointing. It was against this background that the International Health Foundation decided to organize and sponsor their workshop on 'The brain as an endo crine target organ in health and disease' at which the papers appearing in this book were presented.
