Bacterial DNA as well as synthetic singlestranded CpG-DNA oligonucleotides displaying unmethylated CpG motifs activate the vertebrate immune system. Upon engaging toll-like receptor 9 CpG-DNA triggers potent Th1-based immune responses characterized by generation of cytotoxic T lymphocytes (CTL), certain cytokines and antibody subclasses. Here we demonstrate that by covalently linking antigen and CpG-DNA antigen is efficiently directed into intracellular compartments of antigen presenting cells resulting in activation, increased cross-presentation of antigen-derived peptides and thus enhanced induction of antigen-specific CTL responses in vivo. Hence, linked CpG-DNA fulfils a dual role: shutteling antigen-uptake and mediating antigen-specific activation. Since conjugation to stimulatory DNA reduces allergenicity, yet improves immunogenicity of complexed antigens, these conjugates could yield therapeutic potential in vaccine strategies against cancer, allergy or infectious diseases.
