This fourth volume of Contemporary Topics In Immunobiology treats in vertebrate immunity. Specifically, the results represent several approaches to humoral and cellular immunity. It is evident that invertebrates do have function ing immune systems. For example, cellular immunity is characterized by both specificity and memory, but it is still problematical whether vertebrate immune capacity evolved directly from invertebrates. Most of the manuscripts were formally presented at the International Symposium on Invertebrate Pathology, University of Minnesota, August 1972, held in connection with the 25th anniversary celebration of the American Insti tute of Biological Sciences. I wish to express my appreciation to the contributors and to beg their indulgence in what may have been overzealous editing. This was done, though, in the interest of clarity and to seek uniformity. This volume presents a collection of reviews derived from work presented at the Aegean Conference: “4th Crossroads between innate and adaptive immunity”. This meeting was the fourth in a series, and assembled a team of scientists working on mechanisms by which the innate immune system of the host senses pathogens, the cellular and signaling networks that orchestrate the innate response and antigen presentation and adaptive immunity. The importance of the crosstalk between innate immunity and the adaptive immune response has only recently started to be appreciated. Although it is well recognized that dendritic cells, NK cells, NK-T cells and T cells are all critical for the host response to pathogens, the respective fields that study the biology of these immune cells tend to exist in parallel worlds with minimum exchange of information and ideas. This fragmentation hinders the integration of these fields towards a unified theory of host response. The Aegean Conference “Crossroads between Innate and Adaptive Immunity” brought together leading international scientists and experts to address critical areas of Innate and Adaptive immunity something necessary for the development of more efficient scientific exchange and crosspollination between these fields. This conference attracted scientists from all over the world to discuss their latest findings on the various aspects of Innate and Adaptive immunity. The conference had limited participation and a scientific and social program that maximized scientific interchange through lecture presentations, poster sessions and informal discussions. ​

This symposium was established in 1976 for the purpose of bringing to gether once every two or three years, active investigators in the fore front of contemporary immunology, to present their findings and to discuss their significance in the light of current concepts and to identify important new directions of investigation. The founding of the symposium was stimulated by the achievement of major breakthroughs in the under standing of the immune recognition of proteins and peptides. We believed that these breakthroughs will lead to the creation of a new generation of peptides which should have enormous potential in biological, therapeutic and basic applications. This anticipated explosion has finally occurred and many applications of these peptides are now being realized. The main symposia topics of the fourth symposium were: T-cell recognition of proteins, structure and function of the T-cell receptor, presentation of protein antigens, recycling and activation of membrane receptor molecules, Ir-gene control of T-cell responses and methods of cell separation. The molecular features recognized by antibodies on proteins were the first immune recognition sites to be local ized and confirmed by synthetic peptides. The complete antigenic structures of several proteins have been defined, and individual antigenic sites have been described on many more proteins. More recently, major breakthroughs have been reported in the immune recognition of proteins by T cells.

There are many unanswered questions regarding the molecular nature of antibodies, components of complement, and other substances which participate in the immune response. The list of substances which need to be analyzed chemically is increasing. Plasma cell products, of course, have long been of great interest because the most prevalent ones are immunoglobulins. Other cell types, however, are the source of the broad spectrum of additional substances which classically fall into the sanctum of the molecular immunologist. It is these substances, and especially those more recently discovered, which are responsible for the broadening investigative interests of immunologists. In this volume we have provided you with descriptions of research being done with immunoglobulins and with complement. Additionally, we have in cluded two reports that deal with molecules which are among the more recent acquisitions of the molecular immunologist. The components of complement are known to react in a cascading manner which results in the lysis of cellular antigens. The first step in the classical pathway requires the activation of CI by the antibody-antigen aggregates. This volume of Contemporary Topics in Molecular Immunology begins with the report of Reid and Porter which describes their investigation of the mechanism of activation of C I. Their descriptions of C I q and of the reaction of C I with immunoglobulins are especially intriguing.
