Magnetic nanoparticle-based hyperthermia for cancer treatment is a promising technology, particularly when applied as part of an adjuvant strategy with established cancer therapies (chemotherapy, radiation and surgery). In Europe, early clinical trials are being done using magnetic nanoparticle hyperthermia. The studies presented within this thesis examine several different aspects of the use of magnetic nanoparticles in hyperthermia therapies, particularly as an adjuvant treatment strategy with the chemotherapy cisplatinum. Magnetic nanoparticle (mNP) hyperthermia and microwave (915MHz) based hyperthermia were compared for treatment efficacy using induced murine flank tumors. Treatments were evaluated for normal tissue effects, cytotoxicity and resulting therapeutic ratio. In the case of the microwave comparison, mNPH demonstrated an improvement in treatment efficacy, although below statistical significance. However, there was less normal tissue damage with the mNP therapy. When examined as an adjuvant therapy with cisplatinum, it was determined that there is an enhancement of chemotherapy based therapeutic effect, when combined with magnetic nanoparticle hyperthermia. The importance of nanoparticle incubation, which results in changes in nanopartlcle distribution and cellular uptake, was also investigated. Optimization of nanoparticle distribution is essential for creating controlled and effective treatments. These studies indicated that when nanoparticles were allowed to incubate within the tumors of mice, the heating rate of the mNP was decreased. However, the tumor regrowth delay was similar to that seen with immediately activated nanoparticles. This was true both with and without cisplatinum adjuvant therapy. Systemically delivered nanoparticles for the treatment of metastatic cancer is also an attractive therapeutic approach. Cell pellets of different volumes were utilized to determine if cytotoxicity could be achieved with intracellular nanoparticles alone. Results indicate that a minimum number of cells/thermal mass is necessary to achieve intracellular nanoparticle based cytotoxic hyperthermia. In vivo, the use of cisplatinum to modify the tumor environment was investigated as a means to improve the nanoparticle concentration within the tumor. After pretreatment with cisplatinum, nanoparticle concentrations within the tumor increased as much as 3.1 times.

Magnetic fluid hyperthermia (MFH) uses magnetic nanoparticles (MNP) to transform the energy of an alternating magnetic field into heat via magnetic relaxation. This enables organ-confined cancer treatment by delivering cell-damaging temperatures higher than 43°C inside tumors. However, MNP-cell interaction restricts MNP mobility and causes MNP agglomeration, severely limiting the MNP heating behavior. Based on in vitro experiments, this book addresses the applicability of MFH to pancreatic tumor cells and discusses optimizations of intracellular MNP heating for clinical application. The impact of MNP-cell interaction on heating efficiency is quantified and compared to MNP systems mimicking intracellular conditions. Besides, Monte-Carlo simulation of MNP magnetic relaxation is used to predict optimal field parameters and MNP properties to maximize MFH efficiency under medical constraints.
