Inflammation is important in many diseases, yet it is hard to find current information on the pertinent cellular and molecular mechanisms of inflammation. This book is a current and authoritative review of various aspects of inflammation in mammalian organisms. Basic principles, including regulation by cytokines, lipid mediators, reactive oxygen species and leukocyte recruitment are followed by chapters on integrative aspects of inflammation, such as neutrophil extracellular traps, sepsis and granulomatous inflammation. The acute inflammatory response is the body's first system of alarm signals that are directed toward containment and elimination of microbial invaders. Uncontrolled inflammation has emerged as a pathophysiologic basis for many widely occurring diseases in the general population. This book provides an introduction to the cell types, chemical mediators, and general mechanisms of the host's first response to invasion.

The Third Edition of this classic text has been completely revised and updated and greatly expanded in scope. With three new associate editors and over 20 new chapters, the book provides comprehensive coverage of the inflammatory process, its clinical manifestations, and its pharmacologic modulation. This edition includes vital new information on cytokines and cytokine receptors, signal transduction, mechanisms of cellular adhesion, angiogenesis, and wound healing. The "Clinical Correlates" section features new chapters on autoantibodies, sepsis and septic shock, reperfusion injury, type 1 diabetes, inflammation and cancer, cancer immunology, and induction of inflammation by Helicobacter pylori. The section on therapeutic modulation of inflammation includes new chapters on complement inhibitors, cytokine inhibitors, agents targeting transcription factors, and emerging technologies for the discovery of small-molecule therapeutics. Other new chapters in this section cover second line antirheumatic drugs, tolerance to anti-inflammatory drugs, immunomodulation by intravenous gammaglobulin, and antimicrobial peptides.

This book provides readers with the most up-to-date information on cutting-edge research concerning chronic inflammation. We now know that when inflammation becomes chronic, it acts as a strong disease-promoting factor in a variety of disorders including arteriosclerosis, obesity, cancer, and Alzheimer disease. Chronic inflammation is hence called as the “silent killer”; it upsets the body’s homeostatic mechanism insidiously. In spite of these developments, we know very little about the mechanism underlying chronic inflammation. Particularly, we do not know precisely what induces chronic inflammation or what promotes its prolongation in a spatiotemporal framework. Neither do we have clear knowledge about how chronic inflammation destroys various tissues or how it predisposes individuals to many different diseases. To make the situation worse, we have no effective treatment against chronic inflammation. Since 2010, two major research programs (CREST and PRESTO) aimed at clarifying the mechanisms underlying chronic inflammation were launched in Japan, and investigators of different research areas with a brilliant track record were selected by their research proposals. Subsequently they have made their best efforts to answer the conundrum concerning chronic inflammation. This book is a compendium of such research efforts. In each chapter, the CREST- or PRESTO-funded researchers summarize their original work concerning mechanisms of induction, progression, or resolution underlying chronic inflammation. The most emphasized characteristic is the molecular aspect of chronic inflammation. The book thus presents the most recent progress made in the molecular understanding of chronic inflammation.

Cellular and Molecular Mechanisms of Inflammation: Signal Transduction in Inflammatory Cells, Part A is a collection of papers that discusses the mechanisms of the transduction of signals linking stimulated receptors and cellular function. This book describes the pathways of signal transduction involved in stimulating functions of inflammatory cells connected with host defense and development of inflammatory injury. One paper notes the potential of using fluorescence methodology in analyzing ligand-receptor interactions in living systems during the natural abundance of cell surface receptors. Another paper discusses the structure and function of GTP-binding proteins in neutrophil signal transduction, particularly the role of oligomeric G proteins in signal transduction. One concern in signal transduction research is the physiological significance of the presence of multiple forms of proteins that can have identical functions. One paper reviews phosphatidylcholine breakdown and hormone action in the rat liver, focusing on G proteins and on inositol phospholipid breakdown. This book also discusses calcium translocation in signal transduction, as well as, a novel signal transduction pathway involving phosphatidylinositol 3-kinase. This book can prove beneficial for biochemists, micro-biologists, cellular researchers, and academicians involved in the study of cellular biology, physiology or oncology.

In November 1998 many of the key leaders of new drug discovery for inflammatory diseases gathered at Hershey, Pennsylvania for the 9th International Conference of the Inflammation Research Association. The Conference was held over a five day period and provided a stimulating environment for the open exchange of important advances in basic inflammation research as well as new drug discovery and development. This book encompasses some of the highlights of several presentations made at the Conference. It contains some of the latest and important developments in the field of inflammation research. Topics include the status of eotaxin and chemokines in asthma and allergy, signal transduction and regulation of diverse mediators such as the JNK group of MAP kinases, TNF and IL-1 signaling of NF-kB as well as regulators of AP-1, macrophage metalloproteinases, lymphotoxin and further insights into the role of MCP-1 in disease. Also discussed are drug targets in rheumatoid and osteoarthritis, fibrotic diseases,...
