The scope of the 20th Mosbach Colloquium may be best illustrated by the following notes sent to the speakers when the colloquium was organized. "1) The application of inhibitors in cell biology has led to decisive insight into the organisation of cellular units. The subject should be treated against a background of current aspects of cell biology. In some areas of research, a fairly complete picture of the functions and cooperative interactions of the units has already emerged. We will discuss mainly these areas. 2) At this colloquium we want to contribute illustrations of the useful application of inhibitors to biological problems. Due to limited knowledge, inhibitors are sometimes incorrectly employed. This applies both to the planning of investigations and to the con· clusions drawn ("use of inhibitors by uninhibited workers"). 3) Inhibitors themselves are interesting substances and their mechanisms of action constitute fascinating problems." The colloquium has been subdivided into five sections. The known chemical structures of the inhibitors discussed are given in an Appendix. We gratefully appreciate the cooperation of the speakers. To a great extent, they managed the coordination of contributions to each of the five subdivisions. We regret that no contributors from Eastern Europe were able to participate.

During the last decade physical and chemical methods have improved rapidly - a fact which allowed the mode of action of antibiotics to be studied - and many biochemically-oriented scientists have devoted their research to the following questions: 1. What is the metabolic pathway that is inhibited selectively, and what are the target molecules within a sensitive cell? 2. What are the relationships between the chemical structure of an antibiotic and the physicochemical properties of the sensitive mole cule(s)? 3. Why and how far is the action selective? 4. Is it possible to correlate the interaction with the target mole cule(s) with the particular biological activities observed? This monograph deals with those antibiotics which interfere with the biosynthesis of nucleic acids. The idea was to provide an insight into how to investigate the preceding questions experimentally and to solve as yet unresolved problems rather than to give a review of the current state of knowledge. Although the biochemistry of nucleic acid synthesis is known in general, the precise molecular mechanisms by which deoxyribonucleic acid is replicated or transcribed has still to be clarified. For this reason it is not yet possible to describe the molecular mechanisms by which the inhibitors of nucleic acid and protein synthesis exhibit their effects. The fact that the inhibitors of nucleic acid and protein synthesis themselves served as useful tools to obtain an insight into the mechanisms of replication, transcription and translation was one of the most exciting discoveries in this field.

These proceedings contain the papers presented at the Bayer-Symposium V which was held in Grosse Ledder on October 16-21, 1973 and which was simultaneously the Second International Research Conference on Proteinase Inhibitors. The cordial atmosphere, the frank discussions and the profitable exchange of ideas during the First International Research Conference on Proteinase Inhibitors which was held on November 4--6, 1970 in Munich were a stimulant for us to consider holding a Second Conference especially as the steadily increasing research activities in this and related fields called for a new assessment of recent efforts in proteinase inhibitor research. In the three years since the First Conference was held, this field has developed particularly in the areas of molecular aspects of protein-protein interactions and in the extension to inhibitors of nonserine proteinases and peptidases. In planning this meeting the Organizing Committee was faced with the problem of selecting a limited number of participants and we decided that it was appropriate and necessary to focus in detail on the chemistry of proteinase inhibitors, their interac tions with a variety of enzymes and also to invite more general papers to survey the physiological and clinical aspects of the subject which we felt would be of importance in the future.
