Research Paper (postgraduate) from the year 2004 in the subject Biology - Genetics / Gene Technology, grade: 1,0, TU Bergakademie Freiberg, 34 entries in the bibliography, language: English, abstract: Eukaryotic DNA replication depends on the origin recognition complex (ORC) which is conserved from yeast to humans. Activity of replication factors including ORC is coordinated with the cell cycle progression to ensure that the entire genome is copied exactly once per cell cycle. This coordination depends on Cdc2 (cell division cycle 2), also highly conserved across all eukaryotes, which binds to the origin recognition complex. This book presents research results of the chromatin cross-linking immuno-precipitation (ChIP) procedure to determine (1) if Cdc2 kinase is localized at DNA replication origin as predicted by current models, (2) when this happens and (3) what the genetic requirements for that localization are. Furthermore the binding ability of different ORC and cdc2 mutants was tested, as well as the influence of Cdc18 on the ORC-Cdc2 interaction. This is particularly interesting, since those regulations ensure genome stability. Studying these pathways in yeast gives us insights into the mechanisms that are critical in mammalian cells to prevent cancer.

The fission yeast Schizosaccharomyces pombe is the favoured tool of many productive research groups throughout the world, serving as a useful model for fundamental principles and mechanisms, such as genome organization, differential gene regulation, cell-cycle control, signal transduction, or cellular morphogenesis. This book collates the current state of knowledge derived from molecular studies in this simple eukaryotic microorganism. The entire sequence of its genome has been completed, emphasizing the comparative value and model status of this yeast. The individual chapters, highlighting up-to-date views on prominent aspects of molecular organization, were written by active research scientists, presenting the results of their investigations to other workers in neighbouring fields. This book intends to serve the fission yeast community as a handy source of reference for years to come. It will also be of particular value to the ever-increasing number of researchers starting to look into fission yeast affairs for comparative reasons from other platforms of molecular genetics and cell biology.

Nuclear Architecture And Dynamics written by Christophe Lavelle and has been published by Academic Press this book supported file pdf, txt, epub, kindle and other format this book has been release on 2017-10-27 with Medical categories.

Nuclear Architecture and Dynamics provides a definitive resource for (bio)physicists and molecular and cellular biologists whose research involves an understanding of the organization of the genome and the mechanisms of its proper reading, maintenance, and replication by the cell. This book brings together the biochemical and physical characteristics of genome organization, providing a relevant framework in which to interpret the control of gene expression and cell differentiation. It includes work from a group of international experts, including biologists, physicists, mathematicians, and bioinformaticians who have come together for a comprehensive presentation of the current developments in the nuclear dynamics and architecture field. The book provides the uninitiated with an entry point to a highly dynamic, but complex issue, and the expert with an opportunity to have a fresh look at the viewpoints advocated by researchers from different disciplines. Highlights the link between the (bio)chemistry and the (bio)physics of chromatin Deciphers the complex interplay between numerous biochemical factors at task in the nucleus and the physical state of chromatin Provides a collective view of the field by a large, diverse group of authors with both physics and biology backgrounds

Chromatin Regulation and Dynamics integrates knowledge on the dynamic regulation of primary chromatin fiber with the 3D nuclear architecture, then connects related processes to circadian regulation of cellular metabolic states, representing a paradigm of adaptation to environmental changes. The final chapters discuss the many ways chromatin dynamics can synergize to fundamentally contribute to the development of complex diseases. Chromatin dynamics, which is strategically positioned at the gene-environment interface, is at the core of disease development. As such, Chromatin Regulation and Dynamics, part of the Translational Epigenetics series, facilitates the flow of information between research areas such as chromatin regulation, developmental biology, and epidemiology by focusing on recent findings of the fast-moving field of chromatin regulation. Presents and discusses novel principles of chromatin regulation and dynamics with a cross-disciplinary perspective Promotes crosstalk between basic sciences and their applications in medicine Provides a framework for future studies on complex diseases by integrating various aspects of chromatin biology with cellular metabolic states, with an emphasis on the dynamic nature of chromatin and stochastic principles Integrates knowledge on the dynamic regulation of primary chromatin fiber with 3D nuclear architecture, then connects related processes to circadian regulation of cellular metabolic states, representing a paradigm of adaptation to environmental changes

This volume of Advances in Cancer Research begins with a "Foundations in Cancer Research" articles by Harold Varmus. He focuses on Andrew Lwoff who influenced a generation of scientists and how Dr. Lwoff's influence on Howard Temlin, in particular, led to the identification of the cause of AIDS. Hiroto Okayama and colleagues discuss the conserved control mechanisms of the G1 and G2 phases in fission yeasts and mammals, and the newly identified control genes. Nilis Mandahl presents the cytogenetic findings in bone and soft tissue tumors and introduces the major molecular genetic findings. Hannel Tapiovaara dn co-workers review plasmin generation at restricted areas of the cell surface and hypothesize that it may be a catalyst for tumor cells to metastasize. Noël Bouck et al. review the evidence suggesting that certain types of stimulations of inducers by activated oncogenes, and decreased production of inhibitors of angiogenesis, may be instrumental in enabling developing tumour cells to attract new cells and continue the malignant growth. Peter L. Stern reviews the role of immunity and the prospects for immune intervention in cervical neoplasia. Lastly, Denis J. Moss and his associates discuss the Epstein-Barr virus (EBV) host-virus relationship and the immune control of EBV infections and examine development of vaccines and immunotherapy.
