This volume provides a comprehensive and up-to-date overview of the various aspects of Lassa virus (LASV), the causative agent of Lassa fever, a frequently fatal zoonotic disease endemic to West Africa. Compiled by world-leading experts in the field, this book provides an introduction to the 50-year history of Lassa fever research, the genetic diversity of LASV, and its geographic distribution. Subsequent chapters focus on the ecology and epidemiology of LASV, discussing in-depth the interactions of the virus with its hosts - multimammate mice and other rodents - and the interactions between these animal reservoirs and humans. Lassa virus causes Lassa fever disease in several countries in West Africa, where it is estimated to infect up to half million people causing roughly five thousand deaths each year. This deadly virus has also been introduced in multiple occasions into the western world, including the United States, United Kingdom, Netherlands, Sweden, and Germany. Lassa virus infection, which is often misdiagnosed, can lead to a wide range of disease symptoms ranging from mild flu-like symptoms to bleeding disorders, multi-organ failure, and death. Despite some major discoveries made in recent years of research on Lassa fever, there are still many unresolved key issues that hamper the development of effective vaccines and therapies. Some of these issues include a detailed understanding of the viral and participating host factors in completing the virus life cycle, in mediating disease pathogenesis or protection, and in activating or suppressing host immune responses against virus infection. This book is devoted to understanding some of these important issues. OLDSTONE Viruses are generally studied either because they cause significant human, animal or plant disease or for their utility as materials to probe a basic phenomenon in biology, chemistry, genetics or molecular biology. Arenaviruses are unusually interesting in that they occupy both of these categories. Arenaviruses cause severe human diseases known primarily as the hemor rhagic fevers occurring in South and Latin America (Bolivia: Machupo virus and Argentina: Junin virus) and in Africa (Lassa virus). Because such viruses produce profound disability and may kill the persons they infect, they are a source of economic hardship in the countries where they are prevalent. Further, they provide new problems for health care personnel owing to the narrowing of the world as visitors from many countries increasingly travel to and from these endemic areas. In addition, lymphocytic choriomeningitis virus (LCMV) can infect humans worldwide, although the illness is most often less disabling than those elicited by other arenaviruses. Yet LCMV is likely of greater concern to non-arena-virologists and experimentalists using tissue culture or animals, i. e. , workers in molecular biology, cancer research, virology, immunobiology, etc. , because normal appearing cultured cells or tissues and animals used for research may be persistently infected with LCMV without manifesting clinical disease or cytopathology and transmit that infection to laboratory workers (reviewed OLDSTONE and PETERS 1978).
