For the past 30 years I have been teaching lipid biochemistry to'inedical students, graduate students, and undergraduate students. The major topics covered in my courses were fatty acids, prostaglandins, leukotrienes, phospholipids, glyco lipids, triacylglycerols, cholesterol, bile acids, and plasma lipoproteins. Empha sis was placed on the regulation and disorders of lipid metabolism. The latter included hyperlipidemias, atherosclerosis, and alcohol-induced liver damage. In this volume, I have chosen to focus on the disorders of lipid metabolism at a level appropriate both for medical students and for graduate and undergradu ate students majoring in the biological sciences. The biochemistry, nutrition, genetics, and cell biology aspects of lipids and lipid metabolism will be covered as they relate to lipid disorders. I am not aware of any textbook that integrates the disorders of lipid metabolism in this manner. Chapter 1 includes a brief discussion of the basic structures, properties, and metabolism of lipids. This chapter is not very detailed, since the material covered is available in basic textbooks on biochemistry. The major fpcus of this volume is the various lipid disorders, with emphasis on polyunsaturated fatty acids, the molecular biology and pathogenesis of the hyperlipidemias, dietary and drug therapy for the hyperlipidemias, and alcohol-induced liver damage. The material presented has been obtained from several textbooks on biochemistry and from a variety of recent articles in the scientific literature.

This book was stimulated by the enthusiasm shown by attendees at the meetings in Saxon River, VT, sponsored by the Federation ofAmerican Societies for Experimental Biology (FASEB), on the subject of the intestinal processing of lipids. When these meetings were first started in 1990, the original organizers, two of whom are editors ofthis volume (CMM and PT), had two major goals. The first was to bring together a diverse group ofinvestiga tors who had the common goal of gaining a better understanding of how the intestine ab sorbs lipids. The second was to stimulate the interest of younger individuals whom we wished to recruit into what we believed was an exciting and fruitful area ofresearch. Since that time, the field has opened up considerably with new questions being asked and new an swers obtained, suggesting that our original goals for the meetings were being met. In the same spirit, it occurred to us that there has not been a recentbook that draws to gethermuch ofthe informationavailableconcerninghow the intestineprocesses lipids. This book is intended to reach investigators with an interest in this area and their pre- and post doctoral students. The chapters are written by individuals who have a long-term interest in the areas about which they write, and many have been speakers at the subsequent FASEB conferences that have followed on the first.

During the past decade we have witnessed a vast expansion in our knowl edge of lipid metabolism, especially for mammalian tissues. One obvious conclusion arising from these studies is that no single overall scheme of lipid metabolism can be classed as distinctly characteristic of all mamma lian organs. Although certain synthetic and degradative lipid pathways are similar in a variety of organs, I have been impressed by the notable exceptions. I was motivated to organize this work on Lipid Metabolism in Mammals because of the lack of a single reference source containing a comparative organ approach to lipid metabolism in mammals that empha sizes the uniqueness of pathways in the various organs of the body. Because of the escalation in lipid research, I also feel strongly that there is an urgent need for an updated concise account of this field. The group of authors for the chapters in the two volumes of Lipid Metabolism in Mammals were selected for their expertise and personal experience with the lipid metabolism of the organs or blood constituents that are the subjects of the chapters. Sufficient leeway has been given each author to approach the subject matter from a personal viewpoint. How ever, the overall direction of each chapter has been slanted to emphasize the similarities and differences in lipid metabolism among organ systems. The introductory chapter on general pathways provides a convenient refer ence to illustrations of specific reaction sequences that are well established and that occur in a number of organs.

Modification of Lipid Metabolism is a collection of proceedings presented at the Symposium on Modification of Lipid Metabolism, held during the American Chemical Society meeting in Atlantic City, New Jersey, on September 10-11, 1974.
