This book offers a comprehensive review of Lynch Syndrome (LS), addressing both the basic and clinical aspects of this condition. Due to the recent advances in our understanding of the genetic mechanism of LS, and to new screening methods, including universal screening and/or multi-gene panel analysis, the standard treatment strategy for patients and family members of LS have been steadily improving. In this book, experts describe the disease’s manifestations, discuss state-of-the-art diagnosis and management options, and offer a cutting-edge overview of the genetic and epigenetic basis of the syndrome. Providing essential insights into this new phase in the management of LS, this book is a valuable resource not only for colorectal surgeons, but also for general gastrointestinal clinicians, gynecologists, oncologists and all basic researchers with an interest in LS.

Lynch syndrome (LS) is the most common cause of inherited colorectal cancer, a disease with a high mortality rate. An estimated 37,000 of diagnosed colorectal cancer cases worldwide are attributed to Lynch syndrome each year. Intensive cancer screening, with early initiation and frequent follow-up, can reduce colorectal cancer incidence and mortality in LS patients. This book provides an up-to-date overview on the genetic and epigenetic basis of Lynch syndrome. It evaluates clinical features of the disease and critically comments on molecular tools available for identifying mutations responsible for Lynch syndrome; in addition the importance of functional assays that can help clarify the clinical nature of identified mutations is also discussed. The book also focuses on challenges in genetic counselling of at-risk individuals and discusses related ethical issues. The purpose of the book is to give a concise knowledge base for the broader scientific and medical community, including genetic counselors, in order to improve awareness on the potential impact that the diagnosis of LS has on treatment, management and surveillance of LS patients.

It is 1946, and a young man stares out his third-story apartment window. He has returned from the war with metastatic cancer and assumes he will die, leaving his wife and infant daughter behind. Instead, he lives another twenty-four years, raising a family of four children, before he succumbs to a second colon cancer. His son, the author, recognizes that there is a hereditary cancer syndrome in the family and resolves to solve the problem as a medical researcher. Eventually, hereditary colorectal cancer is recognized as a medical entity, and multiple genes responsible for this hereditary condition are isolated. However, the mutation responsible in the author’s family escaped detection. In 2001, his laboratory identifies the mutation responsible for the problem and develops a specific test for the family. This permits the mutation carriers to obtain life-saving care, altering the natural history of the disease for his family and others.

Before My Time written by Ami McKay and has been published by Knopf Canada this book supported file pdf, txt, epub, kindle and other format this book has been release on 2019-09-24 with Biography & Autobiography categories.

Weaving together family history, genetic discovery, and scenes from her life, Ami McKay tells the compelling, true-science story of her own family's unsettling legacy of hereditary cancer while exploring the challenges that come from carrying the mutation that not only killed many people you loved, but might also kill you. The story of Ami McKay's connection to a genetic disorder called Lynch syndrome begins over seventy years before she was born and long before scientists discovered DNA. In 1895 her great-great aunt, Pauline Gross, a seamstress in Ann Arbor, Michigan, confided to a pathology professor at the local university that she expected to die young, like so many others in her family. Rather than dismiss her fears, the pathologist chose to enlist Pauline in the careful tracking of those in her family tree who had died of cancer. Pauline's premonition proved true--she died at 46--but because of her efforts, her family (who the pathologist dubbed 'Family G') would become the longest and most detailed cancer genealogy ever studied in the world. A century after Pauline's confession, researchers would identify the genetic mutation responsible for the family's woes. Now known as Lynch syndrome, the genetic condition predisposes its carriers to several types of cancer, including colorectal, endometrial, ovarian and pancreatic. In 2001, as a young mother with two sons and a keen interest in survival, Ami McKay was among the first to be tested for Lynch syndrome. She had a feeling she'd test positive: her mother's side of the family was riddled with early deaths and her own mother was being treated for the disease. When the test proved her fears true, she began living in "an unsettling state between wellness and cancer," and she's been there ever since. Intimate, candid, and probing, her genetic memoir tells a fascinating story, teasing out the many ways to live with the hand you are dealt.
