Malaria is one of the most widespread and devastating infectious diseases in the world. More than half the world population residing in over 100 countries is at risk of infection from this vector-borne disease. An estimated 250–500 million mal- ial cases occur each year, resulting in nearly one million deaths, the overwhelming majority of which are children. Because of the magnitude of the associated fata- ties, development experts consider malaria a ‘silent tsunami,’ comparing its death toll to the Indian Ocean tsunami (IOT) that ravaged several countries of South and Southeast Asia on December 26, 2004. That tsunami killed some 300,000 people (including children) at once. Globally, malarial deaths account for about 9% of all childhood deaths each year. However, with malaria more than most fatal d- eases, mortality is a small fraction of morbidity. Malaria is a debilitating disease, particularly for the adult population. In addition to children, pregnant women and migrating populations are most v- nerable to malaria. Miscarriage, stillbirth, and low birth weight are common among pregnant women who are infected with this disease. Malaria manifests itself through recurrent fever and chills, with associated symptoms such as anemia and an enlarged spleen. If a person survives the disease, he or she will develop a certain degree of immunity for some years. But malaria victims are not only deprived of energy, they also face an increased risk of other diseases taking hold in the weakened body.

Malaria is present in over 100 countries worldwide, including large areas of South America, Africa and Southeast Asia, and it is estimated that over 40% of the world's population is at risk of infection. The disease is caught through the bite of an infected mosquito carrying plasmodium parasites in its saliva. It is estimated that malaria caused 627,000 deaths in 2012; infections caused by Plasmodium falciparum are the most severe and have the highest mortality rate, but Plasmodium vivax is increasingly recognized as an important cause of clinical symptoms. Antimalarial drugs can be used to prevent and treat malaria, but parasites frequently develop resistance to these agents. Continued research aims to understand the mechanisms of resistance, develop new antimalarial drugs, and identify the best way to prevent being bitten by an infected mosquito. There is currently no licensed vaccine for malaria, although a number of candidate agents are currently being evaluated, and the World Health Organization aims to license vaccines against P. falciparum and P. vivax malaria for use in endemic areas by 2030. Understanding the mechanisms of naturally-acquired malaria immunity is essential to inform vaccine design; many studies are currently underway to explore these immunological mechanisms and identify promising vaccine candidates. This book aims to highlight recent progress in all areas of malaria research, including vaccine development, investigations into new antimalarial agents, vector control and disease epidemiology.

The detection of strains of Plasmodium falciparum in SE Asia which are resistant to chloroquine and other synthetic antimalarial drugs has intensified research on all phases of malaria, including study of the Anopheles fauna of the area. In late 1965 the U.S. Army experienced a severe outbreak of malaria among troops operating in forested areas of Vietnam which were very similar to jungle habitats which have been investigated by entomologists of the U.S. Medical Research and Development Command in neighboring Thailand since 1963. In the latter areas, and presumably in much of Vietnam, Anopheles minimus and A. balabacensis are the most important vectors. The latter species is a particularly effective malaria vector in forested areas in much of SE Asia, but its habits were poorly known until recently. It is highly anthropophilic and bites freely in the jungle. Almost all of the personnel engaged in the investigations in Thailand contracted falciparum malaria despite prophylactic drugs and other protective measures.
