Mechanisms of Cytotoxicity by NK Cells is the resulting book of a workshop focused on the studies that enhance understanding of the basic mechanisms involved in the function and regulation of NK cells. The book presents the status of knowledge of the mechanisms responsible for the cytotoxic effects of NK cells and the regulation of their activity. The text is further divided into five major sections according to the sequence of the workshop sessions. A separate chapter from the sections has been included at the beginning of the book. The First International Workshop on Mechanisms in Cell-Mediated Cytoxicity was held at Carry-Ie-Rouet, France, September 14-16, 1981. The Workshop brought together for the first time leading investiga tors in each of the principal areas of cell-mediated cytotoxicity, as well as experts in the area of complement-mediated cytoxicity. Formal research presentations were held to a minimum, the emphasis being on open discussion of current knowledge about mechanisms of cytoxicity in each of the systems under consideration. The major objectives of the Workshop were 1) to compare and integrate what is known about the mechanism(s) of cytoxicity in each system; 2) to determine whether, on the basis of information in hand, it seems likely that the mechanisms of cytotoxicity in the various systems are the same or are unique; and 3) to stimulate thinking about new approaches to elucidating the fundamental mechanisms by which certain cells are able to kill other cells.

This book is derived from contributions to the Second International Workshop on Mechanisms in Cell-Mediated Cytotoxicity, held in Annopolis, Maryland, June 10-13, 1984. This workshop was organized by an international committee of immunologists interested in lymphocyte cytotoxic mechanisms (G. Berke, W.R. Clark, P. Golstein, M. Hanna, P. Henkart, R. Herberman, H.R. MacDonald, E. Martz, and C. Nathan), who strove to invite participants who have made major contributions to this field. The Workshop was a follow-up to the highly successful 1981 Workshop, whose proceedings Workshop were published by Plenum as Mechanisms in Cell-Mediated Cytotoxicity, edited by W.R. Clark and P. Golstein. That volume has been much appreciated by researchers and students since it contains accounts of most of the current approaches to understanding cytotoxic lymphocyte mechanisms all in one volume. The present book may be viewed as a follow-up to the first one, and in our opinion fairly summarizes the varying current viewpoints on lymphocyte cytotoxic mechanism. It should be noted that the discussions have been transcribed directly by us, and the participants have not had an opportunity to edit their remarks. We have tried to maintain some of the style of the actual discussion in these transcripts. In some cases technical problems prevented usable transcriptions from being made, and hence not all of the actual discussion at the workshop is reproduced here.

During the past 5 years rapid progress has been made in the understanding of biochemical pathways for signal transduction in lymphocyte activation. Gene cloning technology has been instrumental in defining and making available in pure form of a number of growth and differentiation factors, in the characterization of their receptors, and in the delineation of genes for the T cell receptor. This book is divided into 6 sections. Section 1 deals with the molecular structure of the T cell receptor. Section 2 discusses the role of the T cell receptor, membrane ion channels and biochemical pathways of signal transduction in T cell activation. The molecular structures and biological and immunological effects of interleukin 1, interleukin 2 and interleukin 3 are presented in Section 3. This section also details the structure of interleukin 2 receptor and its use as a target for therapy for certain leukemias. Section 4 includes the biochemical events which occur following the delivery of the signal for B cell activation, proliferation, and differentiation by antigen, growth/differentiation factors. The molecular structure of B cell stimulating factors is also discussed. The role of oncogene expression in cellular activation and differentiation is included in Section 5. The cellular and molecular basis of natural killing and the molecular basis of cyc1osporin A-mediated immunosuppression are discussed in detail in Section 6. We hope this book will serve as a reference work on basic mechanisms of lymphocyte activation, proliferation, and differentiation for immunologists and molecular biologists.
