This volume of Progress in Inflammation Research is a unique compilation of work performed by a wide spectrum of investigators from different medical disciplines. It is fascinating that dietary alterations of fatty acid intake can result in a range of salutory changes in a great variety of medical conditions. Most of the good scien tific work which has led to these observations has been performed over just the last two decades. This is of course not a very long time in the context of the history of the human species. Recently performed analysis of fat intake from paleolithic times has indicated that our hunter-gatherer ancestors consumed as much cholesterol as modern Western man, but strikingly less saturated fatty acid and more polyunsatu rates, including n-3 fatty acids. Wild game has the terrestrial source of n-3 incorpo rated in its fat since browsing animals derive 18:3n-3 (alpha-linolenic acid) natural ly from leafy plants. There is, however, little opportunity for modern Western man to get n-3 fatty acids from the diet if one does not consume fish. Modern agribusiness provides ani mal feeds high in n-6 fatty acids, mostly derived from linoleic acid (18:2n-6) in corn feed. Therefore, grazing animals have no access to alternative fatty acids in either feed or grasses, the latter containing little or none of these potentially beneficial highly polyunsaturated fatty acids.

The role of dietary fatty acid (FA) composition in modulating inflammation has b een extensively studied. However, studies were limited to food-derived FAs, rath er than those reported to occur in certain plants, such as members of the Ranunc ulaceae family. The purpose of this study is to investigate potential anti-infla mmatory bio-activities in Ranunculus constantinopolitanus (MP), traditionally us ed in Lebanese folk medicine for the treatment of fungal infections, to compare such activities to those of fish oil and conjugated linoleic acid (CLA), via in- vitro and in-vivo models of inflammation, and to identify the bio-active molecul es underlying these activities. The endotoxin (ET)-stimulated mouse mammary epit helial SCp2 cells were utilized to assess the effect of MP on inflammation marke rs in-vitro. Wild MPI. 2 (wMPI. 2), the chloroform fraction derived from the crude methanol extract of the wild plant, in addition to wild Y2+3 (wY2+3), a FA mix isolated from this fraction and containing palmitic acid, C18:2 and C18:1, presu med to be linoleic and oleic acid, respectively, and stearic acid (1:5:8:1 ratio), reduced ET-induced interleukin (IL)-6 levels, as shown by Enzyme-Linked Immun oSorbent Assay (ELISA), without affecting cell viability or morphology. Our stud y also demonstrates that the observed effect of wY2+3 was not due to any of its individual FA constituents. Furthermore, Electrophoretic Mobility Shift Assay (E MSA) showed that the reduction in IL-6 levels was due to an upstream inhibition of ET-induced nuclear factor-KB (NF-KB) DNA-binding activity by wY2+3. However, wY2+3 did not reduce the activities of ET-induced or serum-derived gelatinase A and B, as monitored by zymography. When compared to fish oil, CLA and an analogo us synthetic FA mix, wY2+3 exhibited higher potency in reducing ET-induced IL-6 levels, both at lower effective concentrations and within a shorter period of ti me. Similar effects to those of wY2+3 were noted in cultivated Y2+3 (cY2+3), tha t showed similarities in their infrared (IR) spectra. cY2+3 did not reverse infl ammation in-vivo. A single intraperitoneal (i.p.) injection of rats with cY2+3 a t 12 mg/kg was not enough to reduce ET-induced paw edema. Lipids are functionally versatile molecules. They have evolved from relatively simple hydrocarbons that serve as depot storages of metabolites and barriers to the permeation of solutes into complex compounds that perform a variety of signalling functions in higher organisms. This volume is devoted to the polar lipids and their constituents. We have omitted the neutral lipids like fats and oils because their function is generally to act as deposits of metabolizable substrates. The sterols are also outside the scope of the present volume and the reader is referred to volume 28 of this series which is the subject of cholesterol. The polar lipids are comprised of fatty acids attached to either glycerol or sphingosine. The fatty acids themselves constitute an important reservoir of substrates for conversion into families of signalling and modulating molecules including the eicosanoids amongst which are the prostaglandins, thromboxanes and leucotrienes. The way fatty acid metabolism is regulated in the liver and how fatty acids are desaturated are subjects considered in the first part of this volume. This section also deals with the modulation of protein function and inflammation by unsaturated fatty acids and their derivatives. New insights into the role of fatty acid synthesis and eicosenoid function in tumour progression and metastasis are presented.
