The Golgi apparatus (GA) is typically comprised of a series of five to eight cup-shaped, membrane-covered sacs called cisternae that look something like a stack of deflated balloons. The GA is often considered the "distribution and shipping department" for the cell's chemical products. This book traces the first 100 years of GA discovery from the first published accounts from Pavia, Italy, in 1898 to the Centenary Celebration in Pavia, Italy, in 1998 to our most recent discoveries. It summarizes the past 50 years beginning with the modern era of GA discovery, leading up to the present era with almost exclusive focus on molecular biology.

In 1898 Camillo Golgi reported his newly observed intracellular structure, the apparato reticolare interno, now universally known as the Golgi Apparatus. The method he used was an ingenious histological technique (La reazione nera) which brought him fame for the discovery of neuronal networks and culminated in the award of the Nobel Prize for Physiology and Medicine in 1906. This technique, however, was not easily reproducible and led to a long-lasting controversy about the reality of the Golgi apparatus. Its identification as a ubiquitous organelle by electron microscopy turned out to be the breakthrough and incited an enormous wave of interest in this organelle at the end of the sixties. In recent years immunochemical techniques and molecular cloning approaches opened up new avenues and led to an ongoing resurgence of interest. The role of the Golgi apparatus in modifying, broadening and refining the structural information conferred by transcription/translation is now generally accepted but still incompletely understood. During the coming years, this topic certainly will remain center stage in the field of cell biology. The centennial of the discovery of this fascinating organelle prompted us to edit a new comprehensive book on the Golgi apparatus whose complexity necessitated the contributions of leading specialists in this field. This book is aimed at a broad readership of glycobiologists as well as cell and molecular biologists and may also be interesting for advanced students of biology and life sciences.

The Golgi apparatus is an organelle found in most eukaryotic cells. The primary function of the Golgi apparatus is to process and package macromolecules, such as proteins and lipids, after their synthesis and before they make their way to their destination. This book presents topical research data in the study of Golgi apparatus, including Golgi organization and stress sensing; signaling pathways controlling mitotic Golgi breakdown in mammalian cells; the role of Golgi apparatus in the biological mechanisms of hypericin-mediated photodynamic therapy; the role of the Trans-Golgi Network (TGN) in the sorting of nonenzymic lysosomal proteins; and the mechanisms involving the role of Golgi apparatus alteration in neurological disorders triggered by manganese.

Analysis Of Golgi Apparatus During Germ Cell Differentiation And Formation And Function Of The Hermes Body Of Epididymal Sperm written by Elliot Byrne and has been published by this book supported file pdf, txt, epub, kindle and other format this book has been release on 2015 with categories.

"Introduction: At the last step of spermiogenesis, Oko et al. (1993) proposed that unstacked cisternae of the Golgi ribbon organized into a structure previously known as the cytoplasmic droplet, here referred to as the Hermes body. Conserved among all mammalian species, the migration of the Hermes body along the midpiece of sperm in the epididymis is observed to coincide with the initiation of sperm motility. The objective of this study was to compare the most abundant protein constituents of isolated Hermes bodies of epididymal sperm as well as Golgi apparatuses of germ cells in order to provide insight into the formation and function of the Hermes body as well as to ascertain the patterns of expression and differentiation of several Golgi markers during spermatogenesis. Methods: 58 antibodies were selected for immunohistochemical localization (IHC) done on sections of adult male rat testes and epididymides (n=4). Reaction patterns were visualized in all germ cell types across the 14 stages of spermatogenesis, in epididymal sperm throughout the epididymal duct, as well as in all somatic cell types in order to establish each protein's pattern of expression. Results: Each of the 58 antibodies analyzed in this study was investigated as a molecular map of protein expression through the 14 stages of spermatogenesis and for their presence in the Hermes body. Quantitatively, the most abundant Hermes body proteins were those for glucose transport (Glut3) and metabolism (Hexokinase 1), each was localized to the Hermes body by IHC. Actin and myosin were identified to propel the Hermes along the flagellum. In addition, the Hermes body contained proteins characteristic of ER, endosomes, and Golgi apparatus both by proteomics as well as by IHC. In the testis Golgi fraction, the transmembrane 9 family of proteins were prominent and of unknown function along with 200 other proteins. The most abundant protein of this fraction was a novel putative glycosyl transferase (GL54D), an unkown protein but verified by IHC in situ. Golgi proteins conserved between the Hermes and the testis germ cell Golgi apparatus and verified by IHC included the TMED family of transmembrane proteins and COPI/II coatomer proteins. Golgi proteins were also revealed during mitosis, meiosis, acrosome and postacrosomal formation, and Hermes body formation, establishing a protein signature for each during germ cell differentiation. Of the 43 proteins localized to the Hermes body of epididymal sperm, only 31 were maintained down the entire duct, which is explained by the presence of members of the ubiquitin-proteasome system. Discussion: The results of this study reveal that although noted in the Hermes body, Golgi apparatus proteins were not abundant in forming and epididymal Hermes bodies. Hence, the flattened cisternae of the Hermes body may represent a hybrid structure that has evolved to suit the specific needs of the sperm during its maturation. Conclusions: The Hermes body is deduced to compartmentalize glycolysis, cytoskeletal, membrane trafficking, stress response, and protein degradation for the initiation of sperm motility and sperm maturation in the epididymis with its complete protein compendium realized in the step 19 spermatid during Hermes body formation. The Golgi apparatus of germ cells also demonstrates a specific complement of proteins that would serve for the differentiation of germ cells during spermatogenesis." --
