Extraordinary advances have been made in the field of human molecular genetics during the past five years. The ability to amplify a specific region ofDNA a millionfold in a few hours using the polymerase chain reaction has led to the rapid identification of mutations in human disease and of DNA sequence polymorphisms on every human chro- some. DNA fragments of up to 1 megabase in length can now be resolved by pulsed-field gel electrophoresis to create long-range physical maps of important regions of the genome, and can be cloned in the form of yeast artificial chromosomes. The discovery of highly variable "minisatellite" DNA sequences has led to the development of DNA fingerprinting. The application of these techniques to the study of the human genome has culminated in major advances such as the cloning of the cystic fibrosis gene, the construction of genetic linkage maps of each human chro- some, the mapping of many genes responsible for human inherited d- orders, genetic fingerprinting of forensic specimens, and the detection of mutations involved in the development of human tumors. Although many of the new techniques in molecular genetics can be learned relatively easily, it is often difficult for a researcher to obtain all of the relevant information necessary for getting up a technique and applying it successfully. The information available in the research lite- ture often lacks the depth of description that the new user requires.

Cell biology involves a range of techniques for examining how cells function, regulate their own behavior, and interact with their neighbors. This book, the first in a series of five comprehensive methods handbooks, covers key protocols in this dynamic field including cellular organelles, cell growth and division, cell movement, cell signaling, and cell death. Because molecular biology approaches are widely used in cell biology, a few essential techniques from that field are also included.

Standard molecular biology techniques are used widely in biomedical re search and have had a major impact on the understanding of physiologic processes and disease mechanisms. They also play an increasing role in mo lecular genetic diagnosis and new therapeutic approaches. This laboratory manual is designed for researchers who need reliable pro tocols for their laboratorywork. It provides a step by step approach to stan dard methods of molecular biology. A wide range of topics is covered by protocols for immediate application in the laboratory. These include pur ification, isolation, labeling, hybridization, sequencing and enzymatic mod ification ofDNA or RNA, polymerase chain reaction, cloning, libraries, and protein techniques. The manual is based on experience with a practical course in molecular biologytechniques, which was taught bythe authors. Since most methods in molecular biology recapitulate in a test tube mechanisms that occur natu rally in the cell, the first three chapters describe and illustrate basic mechan isms of molecular biology. U nderstanding more about these basic mechan isms will enable the researcher to apply techniques more specifically, based on the knowledge of their potential and limits. W e hope that users find this manual a helpful guide on their road to exciting insights.

Recent advances in genetics over the last quarter of a century, especially in molecular techniques, have dramatically reduced the cost of determining genetic markers and hence opened up a field of research that is increasingly helping to detect, prevent and/or cure many diseases that afflict humans. In Statistical Human Genetics: Methods and Protocols expert researchers in the field describe statistical methods and computer programs in the detail necessary to make them more easily accessible to the beginner analyzing data. Written in the highly successful Methods in Molecular BiologyTM series format, with examples of running the programs and interpreting the program outputs, the chapters include the kind of detailed description and implementation advice that is crucial for getting optimal results from human genetic data collected in the laboratory. Thorough and as much as possible intuitive, Statistical Human Genetics: Methods and Protocols aids scientists in understanding the computer programs and analytical procedures they need to use.

Recent insight into the transcripts generated from the mammalian genome (i.e. the transcriptome) has revealed that transcription is a far more complex phenomenon than previously thought. In RNA: Methods and Protocols, expert researchers provide the procedures and methods used to describe the structure of messenger RNAs and non-coding RNAs that are transcribed by RNA polymerase II as the immediate gene products in mammalian cells. Focused on the structure of the RNA products of “gene X” and the mapping of proteins associated with these RNAs, the volume presents appropriate information for non-specialists in RNA biology. Written in the highly successful Methods in Molecular BiologyTM series format, many chapters contain introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and key tips on troubleshooting and avoiding known pitfalls. Comprehensive and practical, RNA: Methods and Protocols views the transcriptional landscape with an appreciation for the role that proteins play in the processing and interpretation of genetic information in an attempt to further our crucial knowledge of the many products and sophisticated regulatory networks that result from it.
