Methods in Plant Molecular Biology and Biotechnology emphasizes a variety of well-tested methods in plant molecular biology and biotechnology. For each detailed and tested protocol presented, a brief overview of the methodology is provided. This overview considers why the protocol is used, what other comparable methods are available, and what limitations can be expected with the protocol. Other chapters in the book present overviews regarding how to approach particular problems and introduce unique methods - such as how to use computer methodology to study isolated genes. The book will be a practical reference for plant physiologists, plant molecular biologists, phytopathologists, and microbiologists.

Plants are able to respond and adapt to changing environmental and endogenous signals by the induction of the synthesis of specific proteins, acting to modify cellular metabolism. Environmental signals include temperature, anaerobiosis and pathogen attack amongst others, whilst endogenous signals include changes in the level of plant growth regulators. In this 1992 text, leading researchers discuss the role that inducible proteins play in cellular metabolism, and the approaches being used to delineate the molecular events leading to their synthesis. Chapters discuss molecular approaches to the study of gene expression, the identification and characterisation of trans-acting transcription factors and attempts to dissect other parts of the signal transduction pathway by the search for pathway mutants. This review volume will be of great value and interest to final year undergraduates, graduate students and researchers in the fields of plant biochemistry and molecular biology.

Methods in Plant Biochemistry, Volume 1: Plant Phenolics reviews current knowledge about techniques used in the analysis of the biochemistry of plant polyphenols and their importance in the agricultural and food industries. It looks at the application of these techniques in the fractionation of cellular constituents, isolation of enzymes, electrophoretic separation of nucleic acids and proteins, and chromatographic identification of the intermediates and products of cellular metabolism. Organized into 15 chapters, this book opens with an overview of the general procedures and measurement of total phenolics, from detecting phenolic substances in crude plant extracts to determining which classes they belong to and the quantitative estimation of total phenol. The reader is introduced to the chemistry, structural variation, function, and distribution of each class of plant phenolics and, in a few cases where this is practicable, detailed listings of known derivatives are given. Most chapters focus on chromatographic separations and high performance liquid chromatography (HPLC), along with thin layer and paper Rf values with HPLC retention times and NMR spectroscopy. The book also outlines the procedures for the extraction, isolation, separation, and characterization of different classes of phenolic compounds, ranging from phenols and phenolic acids to phenylpropanoids, lignins, stilbenes and phenanthrenes, flavones and flavonols, chalcones and aurones, flavanoids, anthocyanins, biflavanoids, tannins, isoflavanoids, quinones, xanthones, and lichen substances. The book is a valuable resource for students, biochemists, and researchers in the plant sciences.

Methods in Plant Molecular Biology is a lab manual that introduces students to a diversity of molecular techniques needed for experiments with plant cells. Those included have been perfected and are now presented for the first time in a usable and teachable form. Because the manual integrates protein, RNA, and DNA techniques, it will serve students, teachers, and researchers in plant physiology, biophysics, and animal molecular biology who have no previous experience handling recombinant DNA or purified proteins. It can also be used by the established molecular biologist who wishes to utilize the powerful techniques of recombinant DNA to explore the mysteries of the plant kingdom. Eight basic experiments which can be used collectively or individually cover Recombinant Cloning and Screening in E. coli; DNA Sequencing Plant RNA Isolation and in Vitro Translations Plant DNA Isolations and Genomic DNA Southern AnalysisChloroplast Isolation and Protein SynthesisPlant Tissue Culture and Agrobacterium Transformations Experiments that have been student tested for three years Blueprints for setting up gel rigs Comprehensive course schedule outlining individual procedures to be finished in each lab segment Course can be tailored to suit the needs of the individual instructor

Preface: Ten years ago, as a young professor, I set out to teach a new course at Purdue University on plant biochemical and physiological techniques. The course was necessary because many graduate students were not receiving adequate training in experimental technique. In the Agriculture School particularly, students were not gaining sufficeint competence and confidence from formal lecture courses in biology and biochemistry to be able to use modern laboratory equipment and follow biochemical procedure. The intent of the course was to provide these students with a wide range of experiments but not necessarily to include all areas of plant physiology. Molecular Biology of Plants: A Text-Manual is an outgrowth of those years of teaching. The book is not complete in a sense of covering all areas of plant biochemistry and physiology. The experiments deal mainly with areas of research (e.g., nucleotides, proteins, and nucleic acids) that I have been most interested in during my career. I hope the manual will serve two major purposes--that it will provide an adequate selection of experimental procedures for an advanced laboratory course in plant biochemistry--physiology and also provide the serious student with a reference book relating to those special areas covered in the manual. War thanks go to the many students who have taken my course and have contributed to this manual.--Joe H. Cherry.

Practical Applications Of Plant Molecular Biology written by Robert J. Henry and has been published by Garland Science this book supported file pdf, txt, epub, kindle and other format this book has been release on 1997 with Science categories.

Practical Applications of Plant Molecular Biology is an important new title which covers the major techniques and how they are applied to a range of vitally important areas. Divided broadly into four sections, this book covers key subjects including the identification of plants and plant pathogens using molecular techniques, the estimation of genetic variation in plants, the use of molecular markers in plant improvement and the use of plant transformation techniques for the improvement of quality and the introduction of resistance. Also included is a comprehensive listing and description of the most frequently used techniques and a set of appendices covering useful topics of reference for the reader. All undergraduates studying plant sciences, molecular biology, biotechnology and agricultural sciences would benefit from having access to this title as would those studying for upper-level Masters courses concentrating on the disciplines covered. This book also provides an invaluable source of reference for professionals in agriculture, plant breeding, crop protection and improvement, biotechnology and molecular biology.
