Micropropagation has become a reliable and routine approach for large-scale rapid plant multiplication, which is based on plant cell, tissue and organ culture on well defined tissue culture media under aseptic conditions. A lot of research efforts are being made to develop and refine micropropagation methods and culture media for large-scale plant multiplication of several number of plant species. However, many forest and fruit tree species still remain recalcitrant to in vitro culture and require highly specific culture conditions for plant growth and development. The recent challenges on plant cell cycle regulation and the presented potential molecular mechanisms of recalcitrance are providing excellent background for understanding on totipotency and what is more development of micropropagation protocols. For large-scale in vitro plant production the important attributes are the quality, cost effectiveness, maintenance of genetic fidelity, and long-term storage. The need for appropriate in vitro plant regeneration methods for woody plants, including both forest and fruit trees, is still overwhelming in order to overcome problems facing micropropagation such as somaclonal variation, recalcitrant rooting, hyperhydricity, polyphenols, loss of material during hardening and quality of plant material. Moreover, micropropagation may be utilized, in basic research, in production of virus-free planting material, cryopreservation of endangered and elite woody species, applications in tree breeding and reforestation.

This Volume contains the papers presented by twenty-eight invited speakers at the symposium entitled, "Genetic Manipulation of Woody Plants," held at Michigan State University, East Lansing, Michigan, from June 21-25, 1987. Also included are abstracts of contributed poster papers presented during the meeting. That the molecular biology of woody plants is a rapidly expanding field is attested to by the large attendance and high level of enthusiasm generated at the conference. Leading scientists from throughout the world discussed challenging problems and presented new insights into the devel opment of in vitro culture systems, techniques for DNA analysis and manipulation, gene vector systems, and experimental systems that will lead to a clearer understanding of gene expression and regulation for woody plant species. The presence at the conference of both invited speakers and other scientists who work with nonwoody plant species also added depth to the discussions and applicability of the information presented at the conference. The editors want to commend the speakers for their well-organized and informative talks, and feel particularly indebted to the late Dr. Alexander Hollaender and others on the planning committee who assist ed in the selection of the invited speakers. The quality of human life has been maintained and enhanced for generations by the use of trees and their products. In recent years, ever rising human population growth has put a tremendous pressure on trees and tree products; growing awareness of the potential of previously unexploited tree resources; and environmental pollution have both accelerated the development of new technologies for tree propagation, breeding and improvement. Biotechnology of trees may be the answer to solve the problems which can not be solved by conventional breeding methods. The combination of biotechnology and conventional methods such as plant propagation and breeding could become a novel approach to improving and multiplying a large number of the trees and woody plants. So far, plant tissue culture technology has largely been exploited by commercial companies in propagation of ornamentals, especially foliage house plants. Generally, tissue culture of woody plants has been recalcitrant. However, limited success has been achieved in tissue culture of angiosperm and gymnosperm woody plants. A number of recent reports on somatic embryogenesis in woody plants such as Norway spruce (Picea abies), Loblolly pine (Pinus taeda), Sandalwood (Santalum album), Citrus and mango (Mangifera indica), offer a ray of hope for inexpensive clonal propagation for large-scale production of plants or 'emblings' or somatic seedlings; protoplast work; cryopreservation; genetic transformation; and synthetic or artificial or manufactured seed production.

Plant cell and tissue culture is now an established method which can be used for clonal propagation of plant material. While such propagation may be uneconomic for field crops which can be readily propagated from seed, the prospects for clonal propagation of woody perennials are greater. A number of woody plants have been shown to produce multiple shoots and roots in vitro, and such micropropagation methods also include embryo culture and somatic embryogenesis. These methods are now becoming established alongside more traditional methods of macropropagation. This book reviews our current knowledge of a number of aspects of these methods for rapid and mass propagaion of fast-growing woody species, and is based on a conference organized by the Committee on the Application of Science to Agriculture, Forestry and Aquaculture (CASAFA) of the international Council of Scientific Unions. It will therefore be of interest to research workers in plant biotechnology and forestry.
