The focus of this chapter is on the development of miRNA diagnostics and the process for miRNA biomarker development. We discuss pancreatic cancer as a model for the application of miRNA-based tests in different clinical biopsy specimen types. Asuragen’s miRInform® Pancreas test illustrates the development of a laboratory developed test (LDT) that was validated in compliance with Clinical Laboratory Improvement Amendments (CLIA) and College of American Pathologists (CAP) regulations. We provide an overview of work on miRNA-based signatures which is focused on resolving clinical dilemmas in solid and cystic pancreatic lesions. We also review some of the promising miRNA candidates identified in biofluids from other cancer types. These studies highlight the emergence of miRNA-based diagnostic tools which have the potential to impact the clinical management of cancer by guiding early diagnosis, aiding in the assessment of patients’ prognosis, and enabling the identification of novel, more effective therapeutic targets.

"The microRNAs (miRNAs) are small (20-24 nt) sized, single stranded, regulatory RNA molecule, or gene regulators of critical transcriptional or post-transcriptional gene regulation in plants in sequence-specific manner to respond to numerous abiotic stresses and animals, non-coding, highly evolutionarily conserved and widely distributed throughout the plant kingdom. The miRNAs are master regulators of plant growth and development, growth and development attenuation under various environmental stresses by stress-responsive miRNAs and plant stress responses and tolerance. The drought, salinity, heat, cold, UV radiation, heavy metal, pathogens and pests and other microbial infections dismay survival, growth, and development, quality, yield, and production of plants. Stress induced miRNAs down regulate their target mRNAs, whereas, their down regulation leads to accumulation and function of positive regulators, recognizing their roles in stress responses and tolerance. Plant miRNA mediated modifications include overexpression or repression of stress-responsive miRNAs and/or their target complementary or partially complementary gene products, miRNA-resistant target genes, target-mimics and artificial miRNAs. Thus, miRNAs may serve as "genomic gold mines", key novel, potent and potential targets in plant genetic manipulations and miRNA-based biotechnology will enhance plant improvement, and crop plant tolerance to different environmental stresses. Amongst the mammalian producer cell lines, the Chinese hamster ovary (CHO) cell lines are of predominant importance in biopharmaceutical production. Thus, novel factors increasing overall productivity are sought and bear the potential to reduce the unit costs of a production process. Furthermore, the current patent situation for several therapeutic proteins demands innovative tools to at least maintain or preferentially increase the cost-effectiveness of their production processes. In this thesis, hitherto unknown factors were revealed by next generation sequencing of chemically mutated and selected CHO-K1 suspension cell lines. Two factors were proven to improve CHO-based production processes: cgrSnord78 and cgrTtc36. The Cricetulus griseus Ttc36 increases the integral as well as the maximal viable cell density and abolishes the cell-cell aggregation whilst cgrSnord78 improves the specific as well as volumetric productivity without significant impact on cell growth. Based on the present results and discussion, foundations for future research on these functionally unrevealed factors are laid. Hence, this work represents the first step towards the application of the genuine biomolecules cgrTtc36 and cgrSnord78 in biopharmaceutical protein production.
