This book describes molecular processes whose deregulation is important in the formation of tumors. The material is developed from basic cell signaling pathways to their roles in the clinical manifestation of specific cancers. Topics covered include molecular events intrinsic to tumor cells (leading to growth deregulation, extended lifespan, and the ability to invade surrounding tissue), protective mechanisms that prevent transformation (including DNA repair and epigenetic regulation), tumor-host interactions (with the endocrine system, the immune system, and blood vessel formation), and the underlying molecular defects of individual cancers.

Cancer is a major public health problem and much research is being conducted to develop effective treatments for various types of malignancies. In doing so, researchers must have comprehensive knowledge about what causes cancer. This book explains the mechanisms of different types of cancers in twelve chapters organized into three sections on oncogenes, tumor suppressor genes, and viral oncogenes.

"The most engaging and accessible account of cancer biology that makes the link between our understanding of cancer and the development of new therapeutics crystal clear. -- Molecular Biology of Cancer: Mechanisms, Targets, and Therapeutics offers an engaging and manageable route into the complex subject of cancer biology. Using the hallmarks of cancer as a foundation, the book describes the cellular and molecular mechanisms underpinning the transformation of healthy cells into cancer cells. -- after discussing a specific biological hallmark of cancer, each chapter shows how this knowledge can be directly applied to the development of new targeted therapies, giving you a clear appreciation of how the theory translated to tackling the disease. The new edition gives a contemporary account of the field, drawing on the latest research but presenting it in a manner that you will find easy to understand. -- New to this edition: *New full colour diagrams help you visualize key concepts more effectively *Separate chapters for growing areas of cancer biology: Metastasis, Angiogenesis, Infectious Agents and Inflammation, and Technology and Drug and Diagnostics Development *Coverage of range of new topics, including immune checkpoints, studying gene function by CRISPR-Ca9, newly proposed mechanisms for the role of obesity in cancer, non-coding RNAs, and the role of exosomes in intercellular communication *Latest details of newly approved therapeutics" -- from back of book.

Angiogenesis is a multi-stage process that drives the generation of new blood and lymphatic vessels from pre-existing ones. It is highly active during embryogenesis, largely inactive during adulthood but reactivated during wound healing and under a number of pathological conditions including cancer and ocular diseases. In addition to endothelial cells, which line the walls of the vessels, several other cell types (pericytes, macrophages, progenitor cells...) also contribute to angiogenesis. A number of signaling pathways are activated and very finely tune the delicate morphogenetic events that ultimately lead to the formation of stable blood proof neovessels. This book reviews recent advances in our understanding of the molecular and cellular mechanisms of angiogenesis, with a focus on how to integrate these observations into the context of developmental, post-natal and pathological neovascularization. The book was published under the auspices of the French Angiogenesis Society. Most contributors are prominent members of this Society or international researchers who have actively contributed to the Annual Meetings of the Society.

In a simplified form, epigenetics refers to heritable changes in phenotype that are not due to changes in the underlying DNA sequence. In this book, epigenetic mechanisms of regulation and dysregulation in health and disease are explored in great depth. Detailed chapters on epigenetic processes including DNA methylation and chromatin post-translational modifications including potential interventions with DNA methyltransferase inhibitors and histone deacetylase inhibitors are explored in initial chapters. These provide a detailed overview and important background to the entire field. The book is then focussed on epigenetic mechanisms involved in various diseases including anti-inflammatory and autoimmune conditions. Important accounts relating to the effects of epigenetics in metabolic syndrome, cardiovascular disease and asthma are the focus of subsequent chapters. The role of epigenetic dysregulation in malignancy is a current topic of interest and represents an intense field of research. A large component of this book is dedicated to the analysis of aberrant epigenetic processes in carcinogenesis and cancer progression. Further, chapters are focused on emerging cancer prevention using nutritional components and anti-cancer therapies particularly with histone deacetylase inhibitors, which have already been approved for the treatment of cutaneous T-cell lymphoma. The emerging role of nanoparticle preparations, especially in the context of delivering potential epigenetic therapies to target cells in various diseases, is also explored in this book. Overall, this book encompasses a wide range of topics related to epigenetic mechanisms in health and disease and would appeal to anyone with an interest in epigenetics, chromatin biology and emerging epigenetic interventions and therapies.
