This volume represents the first collection of articles contributed by research leaders working on the Myb family of transcriptional regulatory proteins. In more than twenty chapters the authors discuss the range of biological processes and diverse cell types in which Myb proteins operate. Although concentrating on the three vertebrate Myb family members, homologues from lower species are also discussed because of the light they are able to shed on the evolution and function of these proteins. Individual chapters describe the involvement of Myb proteins, in particular c-Myb, in normal and diseased development and function of many tissues including haemopoietic cells, blood vessels, the gastrointestinal tract and the brain. Several chapters explore the mechanistic details of the action of Myb proteins, especially structural features, their interaction with DNA and other regulatory proteins, and the variety of genes that are regulatory targets for this group of transcription factors. Abstract: Transcription factors are emerging as key players in the origin of evolutionary novelties and as powerful tools to manipulating plant metabolism. The number of R2R3 Myb genes have amplified dramatically in the plants, where they are involved in the regulation of plant form and metabolic diversity. However, the function of most plant R2R3 Myb genes remains to be determined, primarily due to the lack of phenotypes of loss-of-function mutants. Here, we have investigated the evolutionary origins of R2R3 Myb genes and started to establish the functions of recently duplicated R2R3 Myb genes in maize through a gain-of-function approach. We have identified discrete evolutionary steps involved in the formation of plant specific R2R3 Myb genes from widely distribute R1R2R3 Myb genes. We have used the Black Mexican Sweet (BMS) maize cell culture system to investigate the consequences on the accumulation of metabolites of expressing the novel R2R3 MYB transcription factor ZmMYB-IF35. We demonstrate that, despite the high amino acid identity in the MYB domain with the P1 regulator of 3-deoxy-flavonoid biosynthesis, ZmMyb-IF35 does not induce the accumulation of flavonoids. However, similar to P1, ZmMyb-IF35 induces the accumulation of ferulic and chlorogenic acids, as well as several other compounds not found in the control BMS lines or in P1-expressing lines, suggesting a promising role of ZmMyb-IF35 for engineering the accumulation of various phenolic compounds. Together, our studies indicate that despite their recent common evolutionary origins, ZmMyb-IF35 and P1 have functionally diverged. Theses studies also suggest a possible correlation between phylogeny and function. Whether this is a general characteristic common to all regulatory genes remains to be determined.

The genus Gossypium produces an important crop plant, cotton. Cotton fibers are single-celled trichomes that develop on the maturing ovule. The commercial quality of these fibers varies greatly within the genus. Such morphological variation may be due to differential expression of the genes responsible for fiber development. It has been proposed that these morphological differences could be related to the evolution of transcription factors and their expression. Six myb transcription factors are expressed during fiber initiation in Gossypium hirsutum. Moreover, some members of the myb transcription factor family have been implicated in trichome development in other plants. The molecular evolution of these six fiber-expressed genes was investigated in three members of the genus Gossypium, as well as the evolution of expression in both diploid and allotetraploid cotton.

Additionally, as modification of lignocellulosic biomass is becoming progressively more imperative in the generation of cost-efficient biofuels, the control of phenylpropanoid biosynthesis plays an important role in the achievement of this goal. We identified two R2R3-MYB transcription factors, ZmMYB40 and ZmMYB95, which potentially present a way of fine-tuning lignin composition in maize cell walls, in combinatorial gene expression control with two negative regulators of the phenylpropanoid pathway, ZmMYB31 and ZmMYB42. Maize plants harboring RNAi constructs against ZmMYB40 display statistically lower expression for a number of pathway genes, further supporting this hypothesis. Further, RNA-Seq of rice plants over-expressing the rice homolog of ZmMYB31 elucidates its transcriptional effect beyond those targets already identified in maize, expanding its possible impact.
