This book addresses the differentiation control of skeletal muscle in different locations of the vertebrate body Particular attention is paid to novel regulatory molecules and signals as well as the heterogeneity of origin that have revealed a developmental overlap between skeletal and cardiac muscle. Different functional muscle groups are the product of the evolution of the vertebrate classes, making a phylogenetic comparison worthwhile for understanding the role of muscle stem cells and precursors in myogenesis. New insights into the hierarchy of transcription factors, particularly in the context of these different muscle groups have been gained from detailed investigations of the spatio-temporal and regulatory relationships derived from mouse and zebrafish genetics and avian microsurgery. Importantly, epigenetic mechanisms that have surfaced recently, in particular the role of MyomiRs, are also surveyed. With an eye to the human patient, encouraging results have been generated that identify parallels between embryonic myogenesis and regenerating myofibers due to common regulatory molecules. On the other hand, both processes differ considerably in quality and complexity of the processes employed. Interestingly, the heterogeneity in embryonic sources from which skeletal muscle groups in the vertebrate including the human body take origin is paralleled by differences in their susceptibility to particular muscle dystrophies as well as by the characteristics of the satellite cells involved in regeneration. The progress that has been made in the field of muscle stem cell biology, with special focus on the satellite cells, is outlined in this book by experts in the field. The authors review recent insights of the heterogeneous nature of these satellite cells regarding their gene signatures and regeneration potential. Furthermore, an improved understanding of muscle stem cells seems only possible when we study the impact of the cell environment on efficient stem cell replacement therapies for muscular dystrophies, putting embryological findings from different vertebrate classes and stem cell approaches into context.

Myogenesis is the formation of adult skeletal muscle from muscle precursor cells. Muscle tissues make up the largest body tissues in an organism. Skeletal muscle is essential for locomotion, and movement and is voluntarily controlled by the organism, unlike cardiac and smooth muscle. It has a highly organized cytoskeletal system. One of the distinct characteristics of skeletal muscle is the striation contributed by the repeating unit of actin and myosin, together they form a single myofiber. This repeating unit of actin and myosin, known as sarcomere, establishes the skeletal muscle unit. If we investigate the developmental origin of skeletal muscle, it starts from the paraxial mesoderm. Paraxial mesoderm is a tissue formed in blastopore or primitive streak during the process of gastrulation. During embryo axis elongation, at the posterior tip of the embryo paraxial mesoderm forms presomitic mesoderm. The anterior part of the transient presomitic mesoderm structure is committed to form somite. It is the somite from where myogenesis starts. Initiated with skeletal myoblast cells, myogenesis involves proliferation and differentiation which ultimately leads to the formation of multinucleated myofibers by the fusion of mononucleated myoblasts and myocytes. A vast number of transcription factors control the process of myogenesis. These factors are known as myogenic regulatory factors (MRFs) and are member of basic helix loop helix family transcription factors. Activation of these MRFs is the hallmark of the initiation of myogenesis. Here we discussed different factors that contributed to the various stages of myogenesis starting from the embryonic stage to tissue formation. In general, muscle development has two major phases, one is embryonic developmental phase from where skeletal muscle tissue develops and another is muscle regeneration which is more prominent in adult muscle tissue after facing injury. Both development and regeneration need regulation by MRFs. The analysis of cell fusion and the mechanism behind it started long back in different systems. Starting from the embryonic stages we have found cell fusion as a normal physiological phenomenon in neural cell, yeast reproduction, exocytosis, macrophage, and muscle cells. Here, we addressed the fusion of mouse myoblasts where two or more myoblast cells fused to form multinucleated myotubes. Apart from the vertebrate system, mechanism of myoblast fusion has also been established in Drosophila and zebrafish systems. Despite the differences in gene expression, the fundamental mechanism of muscle fusion remains the same across systems.

The multi-step process of skeletal muscle development begins with the specification and commitment of precursor cells to the muscle program. Committed cells express the myogenic regulatory transcription factors that control skeletal muscle differentiation. Utilizing the P19 cell system, I have examined the mechanisms by which various factors regulate the expression and activity of the myogenic factors during commitment and differentiation of skeletal muscle. I have shown that the signaling molecule, Wnt3a, acts via nuclear beta-catenin to activate the expression of somite transcription factors including Gli2, Pax3, Six1 and Mox1. beta-catenin was shown to be sufficient and essential for the expression of these somite factors and skeletal muscle development during P19 cell differentiation. Furthermore, I have shown that Gli2 expression induces the expression of Pax4, Mox1 and the myogenic factors leading to skeletal myogenesis. Expression of a dominant negative form of Gli2 indicates that Gli2 activity is necessary for skeletal muscle development. The observation that Pax3 expression could not compensate for the loss of functional Gli2 suggests that the activity of Gli2 and Pax3 during skeletal myogenesis may be interdependent. I also examined the mechanism by which E type binding partners may inhibit activity of the myogenic regulatory factors. Full length forms of E12/E47, as well as ITF-2B, were found to inhibit MyoD activity on exogenous but not endogenous promoters. Therefore, the ability of the E type factors to negatively regulate MyoD activity is contingent on the context of the E box site. In conclusion, these studies further elucidate the mechanisms by which various factors regulate the myogenic regulatory factors during the processes of commitment and differentiation during skeletal muscle development.
