There has been a convergence in recent years of people from the physical and biological sciences and from various engineering disciplines who are interested in analyzing the electrical activity of nerve and muscle quantita tively. Various courses have been established at the graduate level or final-year undergraduate level in many universities to teach this subject matter, yet no satisfactory short text has existed. The present book is an attempt to fill this gap, and arises from my experience in teaching this material over the past fifteen years to students on both sides of the Atlantic. Although covering a wide range of biophysi cal topics from the level of single molecules to that of complex systems, I have attempted to keep the text relatively short by considering only examples of the most general interest. Problems are included whenever possible at the end of each chapter so the reader may test his understand ing of the material presented and consider other examples which have not been included in the text.

One of the most active and productive areas of biological science in the past decade has been the study of the biochemical and biophysical prop erties of cell membranes. There is little doubt that membranes are essen tial components of all cellular systems and that each type of membrane manifests specific and characteristic cellular functions. In the nervous system, important events such as neurotransmission, receptor binding, ion transport, axonal transport, and cell uptake are all known to take place within the neural cell membrane. Phospholipids, one of the major components of membranes, not only provide the membrane with its structural integrity and physical proper ties, but also play an important role in regulating membrane function. Attention has recently been focused on the asymmetric localization of these molecules, the identification of discrete metabolic pools of phospholipids within the membrane matrix, and their involvement in sig nal transmission. Although synaptic membranes generally lack an active mechanism for the de novo biosynthesis of phospholipids, a number of enzymic routes are present for their interconversions and for facilitating metabolic turnover. Metabolites generated during the interconversion reactions may also exert a great influence in modulating membrane func tions. The phosphogylcerides of neural membranes are especially enriched in polyunsaturated fatty acids. However, only very small amounts of these fatty acids are present in the free form, and they are maintained in dynamic equilibrium with the membrane phospholipids.

The Russian edition of this book appeared in 1969 and im mediately gained widespread recognition as a reference work for research workers interested in the physiology, biophysics, and pharmacology of excitable tissues. There are several reasons for the book's success. It deals with a key problem in biology which has recently been the subject of very intensive study and it is of great interest to a wide scientific audience. Not only the fundamentals of the modern membrane theory of biopotentials, but also the vast factual material collected in the last decades by the study of the biophysical and pharmacological properties of the ionic permeability pores of the cell membrane, are described in the book in an authoritative yet readable form. Special attention is paid in the book to the systematic analysis of the consequences of the Hodgkin-Huxley mathematical theory of the nervous impulse for the problem of excitability. The relationship between the various parameters of excitability (threshold potential, threshold current, useful time), accommodation, and the action potential on the one hand, and the constants of ionic permeability of the nerve fiber membrane, on the other hand, is subjected to detailed examination in this context. To do this, the author has made extensive use not only of experimental results obtained on isolated fibers (especially single nodes of Ran vier), but also the results of his own investigations on mathematical models of excitable membranes.

The research on this project was carried out along two closely related lines: One line of investigation was devoted to the theoretical analysis of transport processes across membranes using nonequilibrium thermodynamics. In the framework of this project chemicodiffusional coupling was considered and applied to facilitated or carrier transport. The second line of investigation was essentially experimental in approach. (a) In an attempt to construct a well defined model for very thin biological membranes, the kinetic coefficients for ion permeability through monolayers were determined. The monolayers were adsorbed at the interface between a mercury electrode and water, and the ionic permeability measured polarographically. By this method the transport of Cu(++) across a decylammonium monolayer and its dependence on temperature was investigated. (B) The transport of ions across a composite double membrane was measured as a function of the applied potential. In a membrane composed of cation and anion exchange elements in series, the ionic distribution was studied, and the results analyzed by the methods of nonequilibrium thermodynamics. (Author).
