Metabolic disorder caused by altered levels of metabolism resulting pathophysiological abnormalities often leads to childhood death. Several new developments on metabolic diseases research have been emerging. Gaucher disease is a lysosomal storage disorder caused by glucocerebrosidase gene mutations resulting glucocerebrosidase deficiency. Current studies show that the same gene mutations also contribute to the Parkinson's disease. Tetrahydrobiopterin (BH4) has been widely used in treating patients with Phenylketonuria over a decade. Recent studies reveal that patients treated with BH4 over one-year period showed reduced levels of serum B12, folate and iron intake and therefore patients under chronic BH4 treatment needed to be advised to have additional micronutrients along with BH4. Macrocephaly was used as one of the important features to diagnose Canavan disease. However, a recent study showing a child with aspartoacylase gene mutation developed microcephaly. Hence, Canavan disease phenotype can be either macrocephaly or microcephaly. These are a few examples of recent developments on metabolic diseases research. Therefore, this book was aimed to compose current developments on metabolic diseases research for the use by broad spectrum of experts including Physicians, Neuroscientists, Neurologists, Biomedical researchers, Biochemists, Molecular biologists, Basic Science Researchers and Medical Students.

This book is based on the Symposium "Metabolic Regulation and Functional Activity in the Central Nervous System" which was held on September 16 and 17, 1972, at Saint Vincent (Aosta)/Italy, and was sponsored by the Accademia di Medicina di Torino with the scientific cooperation of the Istituto di Farmacologia, Universita di Torino, and the Pharmakologisches Institut der Freien Universitat Berlin. Its purpose was to give a greater number of scientists from different countries an opportunity to report their latest results under a heading concerning general relationships between metabolism and function. We quite deliberately refrained from pursuing the partly heterogenous subjects into details. Thus, the organizers hoped to be able to interest a greater circle of readers for the manifold subjects from various fields dealing with the investigation of metabolic processes in the central nervous system. The discussion remarks to the lectures could not be considered for printing. The Accademia di Medicina di Torino and all others concerned do not only thank Dr. R. This book is based on an international symposium titled "Cytochrome oxidase in energy metabolism and Alzheimer's disease," held as a satellite to the 27th meeting of the Society for Neuroscience, New Orleans, 1997. The symposium was dedicated in honor of Dr. Margaret T. T. Wong-Riley because, in our opinion, the cytochrome oxidase histo chemical method introduced by Dr. Wong-Riley in 1979 was the most significant break through to map energy metabolism in the entire brain since the 2-deoxyglucose method introduced by Dr. Louis Sokoloff and colleagues in 1977. Both of these metabolic map ping techniques have made monumental contributions to brain research by allowing an integral view of brain activity. They have also developed into various specialized tech niques, including applications to the human brain. One of these new applications, which is described in detail in this book, is the quantitative cytochrome oxidase cytochemical method used to study Alzheimer's disease. The objective of this book is to describe the role of cytochrome oxidase in neuronal metabolism and Alzheimer's disease. Whether genetic or environmental, the pathogenesis of Alzheimer's disease involves a cascade of multiple intracellular events, eventually re sulting in failure of oxidative energy metabolism. Could impairment of cytochrome oxi dase in energy metabolism initiate the degenerative process? Cytochrome oxidase function and dysfunction are discussed in relationship to neuronal energy metabolism, neurodegen eration, and Alzheimer's disease. The book is made up of 10 chapters, divided into three major parts.
