The contributions to this book derived from the Seventh Munich Symposium on Microbiology on June 3 and 4, 1981, which was organized by the WHO Centre for Collection and Evaluation of Data on Comparative Virology at the Institute of Medical Microbiology, Infectious and Epidemic Diseases, University of Munich, Federal Repub lic of Germany. One of our principal purposes was to establish a forum at which the comparative aspects of questions of current interest in the field of medical virology could be discussed. In addition to the presentation of recent fmdings in microbiology, our overall aim was to crystallize trends and indicate new directions for future research activities. This book is a topical review of "New Horizons in Diagnostic Virology." Every one interested in virology is aware of the tremendous progress made in viral diagnostic techniques during recent years and the growing importance of viral diagnosis in human and veterinary medicine. There is yet another step that diagnostic virology has to take: the introduction on a routine basis of methods of molecular biology into the viral diagnostic laboratory. The application of monoclonal antibodies and techniques for the chemical and biological identification of proteins, carbohydrates, and enzymes are discussed, as is the introduction of techniques for the characterization of nucleic acids in viral diag nosis.

The traditional approach to diagnosis of virus infectians by isolation of the causative virus is usually both slow and expensive. More recently, the emphasis has been on the direct detection of viruses or viral antigens in clinical specimens. This can be done using established techniques such as immunofluorescence or electron microscopy, or by newly developed biochemical methods. The purpose of this meeting was to review these and other developments in the laboratory diagnosis of virus infections. The basis of all immunoassays is the interaction of antibodies with antigens. The most widely used immunoassay technique is radioimmunoassay (RIA) which was first developed by Yalow and Berson in 1959. The principle of RIA is elegantly simple. It utilizes a competitve binding reaction between analytes and a radio-labeled analog of the analytes (the tracer) for anti-analyte antibodies. In addition to its exquisite specificity, extraordinary sensitivity, good accuracy and precision, ease and rapidity of assay and simplicity of assay development, the applicability of RIA to a wide variety of substances has made it one of the most powerful and versatile analytical methods of the 20th century and beyond. Millions of RIA's are being performed annually on clinical, biological and environmental samples in licensed laboratories. In order to expand the use of RIA beyond the confines of these laboratories to areas like physician's offices, patients' homes, economically less developed countries, agricultural fields, large scale and continuing screening tests for infectious diseases, it has become necessary to develop non-isotopic labels. Indeed the last fifteen years have seen the development of a great number of ingenious non-isotopic labels in immunoassay so that a whole new industry capitalizing on the potential market for non isotopic immunoassays has appeared. It is the purpose of this volume to present in depth, state-of-the-art reviews on techniques used in non-isotopic immunoassays.
