The last decade has seen remarkable advances in human ge netics. Once the correct chromosome number of the human genome was ascertained, a wide variety of diseases was rec ognized as due to numerical chromosome anomalies. There followed the discovery that spontaneous abortions are the result of chromosome errors, and specific band patterns of chromosomes allowed identification of minute lesions. The techniques of cell hybridization now allow specific gene assign ment to chromosomes and even to distinct loci on their arms. All this was possible because of the ease with which metaphase chromosomes can be obtained and manipulated. The much older technique of analysis of meiotic chromosomes has taken a back seat in this exciting era. Being much less readily accessi ble, spermatogonial analysis is much less frequently under taken and is less successful. Even more difficult for study is the female meiotic process. Not only is meiosis extraordinar ily long, spanning from before birth to ovulation, the tech niques for its study and the patience required for detailed inquiry have been significant obstacles. At the same time, the suspicion that female meiotic analysis should not only be rewarding but that it may be mandatory has been with us ever since it was recognized that a positive correlation exists between chromosomal nondisjunction and maternal age. Before the intricacies of chromosomal behavior that are re sponsible for nondisjunction are understood, however, it is necessary that we comprehend the normalcy of the process.

Studies of oogenesis occupy an important place in current in vestigations in developmental biology. Today no one has any doubt whatsoever that oogenesis is not just the prelude to development, but is development itself, and a very essential part of it. These words of an eminent Soviet scientist, B. L. As taurov , taken by T. B. Aizenshtadt as an epigraph to her chapter in this book, make a good epigraph for the entire book. It is now clear that during oogenesis not only vast reserves of ribosomes and mitochondria, of yolk, carbohydrates, and lipids, and of enzymes for protein and nucleic acid synthesis and for carbohydrate and fat metabolism (which ensures the energy supply and metabolism of the oocyte and the developing embryo) are formed, but also long-lived mRNA and proteins are synthesized, which determine both the completion of oocyte maturation and the initial stages of embryonic development. In the last 15-20 years, the use of molecular biology methods, electron microscopy, autoradiography, and microsurgical methods of experimental embryology in studying the pre-embryonic development of animals has greatly increased our knowledge of oogenesis. This has led to the need to systematize the data obtained, to reinter pret old ideas, and to review the results obtained by new research trends which have emerged in the last few years and which are of general biological interest. Such a task was undertaken in the book Sovremennye Problemy Oogeneza (Current Problems of Oogenesis), published in 1977 (in Russian).

This volume covers the current knowledge base on the role of signaling and environmental pathways that control the normal development of germline stem cells, meiotic progression of oocytes, events of oocyte maturation and fertilization, and the birth of an embryo. Germ cells are uniquely poised to sustain life across generations through the fusion of oocyte and sperm. Because of the central importance of germ cells to life, much work has been dedicated to obtaining a clear understanding of the molecular and signaling events that control their formation and maintenance. Germ cells are set aside from somatic cells in the embryo and go through specialized meiotic cell cycles as the animal matures. These cell cycles are interspersed with long periods of arrest. In human females, meiosis I is initiated in the fetus. At birth, oocytes are arrested in meiosis I; after puberty, every month an oocyte initiates meiosis II – ovulation. Upon sperm availability these cells are fertilized, generate an embryo, and the cycle-of-life continues. During meiotic I progression and arrest, the fitness of oocytes and their progeny are likely influenced by environmental cues and signaling pathways. A lot of recent work has focused on understanding the mechanisms that regulate oocyte fitness and quality in humans and vertebrates. Much of our understanding on the events of meiosis I and germline stem cell populations comes from work in invertebrates, wherein the germline stem cells produce oocytes continuously through adult development. In both inverbrates and vertebrates nutritional and signaling pathways control the regulation of stem cells in such a manner so as to couple production of gametes with the nutritional availability. Additionally, mature oocytes arrest both in meiosis I and meiosis II, and signaling and nutritional pathways have been shown to regulate their formation, and maintenance, such that despite long periods of arrest, the oocyte quality is assured and errors in chromosome segregation and varied cytoplasmic events are minimal.
