The objective of this book is to provide a unifying approach to the study of biophysical chemistry for the advanced undergraduate who has had a year of physics, organic chem istry, calculus, and biology. This book began as a revised edition of Biophysical Chemistry: Molecules to Membranes, which Elizabeth Simons and I coauthored. That short volume was written in an attempt to provide a concise text for a one-semester course in biophysical chemistry at the graduate level. The experience of teaching biophysical chemistry to bi ologically oriented students over the last decade has made it clear that the subject requires a more fundamental text that unifies the many threads of modem science: physics, chem istry, biology, mathematics, and statistics. This book represents that effort. This volume is not a treatment of modem biophysical chemistry with its rich history and many contro versies, although a book on that topic is also needed. The Physical Basis of Biochemistry is an introduction to the philosophy and practice of an interdisciplinary field in which biological systems are explored using the quantitative perspective of the physical scientist. I have three primary objectives in this volume: one, to provide a unifying picture of the interdisciplinary threads from which the tapestry of biophysical studies is woven; two, to provide an insight into the power of the modeling approach to scientific investigation; and three, to communicate a sense of excitement for the activity and wholesome argument that characterize this field of study.

This volume contains the proceedings of the FEBS Sym posium on the Biochemistry of Membrane Transport, which was held at the Swiss Institute of Technology, Zlirich, July 18-23, 1976. Of the speakers invited or iginally, only five could not attend the meeting, and of the lectures given, all but one of the texts are published here. Thus, this volume gives a faithful ac count of the way the meeting was originally conceived and actually took place. This Symposium on Biochemistry of Membrane Transport was the first Symposium sponsored by the FEBS outside the yearly FEBS-Meetings, after the Special Meeting on Industrial Biochemistry, which took place in Dublin in 1973, and it reflects the interest and the trend for gatherings of smaller size than the official FEBS Meetings. The topic of the Symposium was an easy choice, not only because membrane transport is becom ing more and more important to biochemistry every year, but also because of the long-standing interest of Swiss Science in the field. In the choice of the topics and of the speakers, efforts were made to achieve as balanced a coverage of the area as possible. However, since some aspects of membrane biochemistry were dealt with extensively at the parallel 10th Inter national Congress of Biochemistry in Hamburg, GFR, cer tain topics were given less emphasis than others. The Symposium was attended by about 400 participants (we expected 200-250); among them 48 were invited speakers, and some 200 contributed posters.

The strong trend in the Biological Sciences towards a quantitative characterization of processes has promoted an increased use of thermo dynamic reasoning. This development arises not only from the well known power of thermodynamics to predict the direction of chemical change, but also from the realization that knowledge of quantitative thermodynamic parameters provides a deeper understanding of many biochemical problems. The present treatise is concerned primarily with building up a reliable data base, particularly ofbiothermodynamic and related quantities, such as partial specific volumes and compressibilities, which will help scientists in basic and applied research to choose correct data in a special field that may not be their own. Most chapters reflect this emphasis on data provision. However, it was also felt that the expert user deserved information on the basic methodology of data acquisition and on the criteria of data selection. Therefore all tables are preceded by a critical evaluation of the techniques as well as a survey of the pertinent studies in the corresponding areas. The surveys are usually self-consistent and provide references to further sources of data that are important but not covered in the present volume. The reader will realize that in different chapters, different symbols have be~n used for the same properties. This unfortunate situation is particularly obvious in those chapters where partial specific or molar quantities had to be introduced; however, it also occurs in those contributions concerning phase changes of macromolecules.
